高效液相色谱法测定清肝化瘀合剂中异鼠李素含量

摘　　要：目的建立测定清肝化瘀合剂中异鼠李素含量的高效液相色谱(HPLC)法。方法采用高效液相色谱仪,色谱柱为HypersilODS2柱(250mm×4.6mm,5μm),流动相为乙腈-0.8%磷酸水溶液-水(33∶25∶42),流速0.6mL/min,检测波长360nm,柱温25℃。结果异鼠李素质量浓度在0.9429～7.5428μg/mL范围内与峰面积线性关系良好(r=0.9999),平均加样回收率为99.54%,RSD为1.28%(n=6)。结论所用方法简便、快捷,准确度高,可用于清肝化瘀合剂中异鼠李素的含量测定。Objective To establish a HPLC method for determining quantitatively isorhamnetin in Qingganhuayu Mixture.Methods High Penformance Liquid Chromatography was adopted.The column was Hypersil ODS2（250 mm×4.6 mm,5 μm）,the mobile phase was acetonitrile-0.8%phosphoric acid solution-water（33 ∶25 ∶42,V/V）,the flow rate was 0.6 mL/min,the detective wavelength was 360 nm,the column temperature was 25 ℃.Results The linear range of isorhamnetin was 0.942 9-7.542 8 μg/mL（r=0.999 9）,the average recovery rate was 99.54%,and RSD was 1.28%（n=6）.Conclusion The method is simple,rapid and accurate,which can be applied to the determination of isorhamnetin in Qingganhuayu Mixture.
