摘  要:目的:观察运动、注射IGF-1对大鼠骨骼肌内IGF-1含量、mTOR、AR蛋白表达的影响,探讨IGF-1对mTOR、AR的调节作用。方法:雄性成年SD大鼠24只,随机分为4组,安静对照组(C)、安静+IGF-1注射组(CI)、运动组(E)、运动+IGF-1注射组(EI),每组6只。运动方案为60 min/d,强度20 m/min,坡度10%,持续一周的跑台训练。每天训练后大鼠小腿后侧皮下注射IGF-I,对照组注射安慰剂。最后一次运动后6 h取胫骨前肌,测定IGF-1含量、mTOR、AR蛋白表达。结果:1)与C组比较,E、EI组骨骼肌内IGF-1含量、mTOR蛋白表达显著升高;2)C、CI、E、EI组骨骼肌内AR蛋白表达呈下降趋势。结论:1周耐力运动可导致肌肉内IGF-1含量、mTOR蛋白表达显著增加,而肌肉中AR蛋白表达却呈现出下降趋势,其原因有待于进一步研究。The study examined the effects of IGF-1 injection and exercise on the protein expression of mTOR,AR and IGF-1 level in rats skeletal muscle.24 male Sprague-Dawley rats were randomly divided into four groups: inactive control(C), inactive plus IGF-1 injection(CI),exercise(E),exercise plus IGF-1 injection(EI).After all groups had been accustomed for one week,the Group E EI performed closely monitored training in the treadmill(grade of 5,at the speed of 20m/min,duration of 1 hour) for 7 days.When the exercise programs were completed every day,IGF-1 or placebo was injected subcutaneously in the back of right leg immediately.After 6 hours of the last exercise,Tibialis anterior muscle(TA) was isolated and IGF-1 level,the protein expression of mTOR and AR were determined.In our study,exercise evoked a significant(p0.05) increase in IGF-1 level and mTOR protein expression in the TA.Compared with Group C,there was a downtrend in AR protein expression in other three groups,without significant difference(p0.05).The results indicate that one week endurance exercise stimulus induces an increase in IGF-1 level and mTOR protein expression.But the phenomenon of AR decrease should be taken further verification.
