补肾与健脾方对骨质疏松症大鼠干预作用及下丘脑PKC蛋白表达的影响

摘　　要：目的:探索骨质疏松症的病理机制,比较补肾与健脾中药干预骨质疏松症的作用。方法:摘除大鼠卵巢建立骨质疏松症模型,给予补肾、健脾中药灌胃3个月,Western blotting法检测指标。结果:与正常组比较,骨质疏松症模型空白组骨密度明显下降(P<0.01),下丘脑中PKCα与PKCβ2蛋白表达显著增加(P<0.01)。与模型空白组比较,补肾中药高剂量组骨密度显著增加(P<0.01);补肾、健脾中药均可下调模型大鼠下丘脑中PKCα与PKCβ2蛋白表达(P<0.01)。结论:补肾中药防治骨质疏松症的作用优于健脾中药;骨质疏松的发生可能与脑PKC的变化有关,补肾中药对其有调节作用优于健脾中药。Objective:To explore osteoporosis compared with the pathological mechanisms as well as tonifying kidney and tonifying spleen medicine intervention of osteoporosis.Methods:By using ovariectomy,we established the rat model of osteoporosis due to Kidney-deficiency.We had given the factors interfere with ton-ifying kidney and tonifying spleen medicine orally 3 months.Then get the materials and test the index by Western blotting method.Results Compared with the normal group,osteoporosis model blank group decreased bone mineral density(P < 0.01);the protein expression of PKCα and PKCβ2 in hypothalamus increased significantly(P < 0.01).With a blank model group,tonifying kidney medicine(highdose group) can be a significant increase in bone mineral density(P < 0.01),while the role of ton-ifying spleen medicine was not obvious,and tonifying Kidney medicine(highdose group)significantly different(P <0.01);but tonifying Kidney and tonifying spleen medicine can be reduced in the hypothalamus of rat model of PKCα and PKCβ2 pro-tein expression(P < 0.01).Conclusion:The role of tonifying Kidney medicine for prevention and treatment of osteoporosis is better than tonifying spleen medicine;the incidence of osteoporosis may be related to changes in the brain related to PKC,tonifying Kidney,tonifying spleen all of its Chinese medicine with the adjustment on PKC,and the role of tonifying Kidney medicine is better than tonifying Spleen med-icine.
