摘  要:探讨血管紧张素转化酶抑制剂卡托普利对高血压大鼠心肌内质网应激时葡萄糖调节蛋白78((glucose-regulated pro-tein78,GRP78)和C/EBP同源蛋白(C/EBPhomologous protein,CHOP)的表达及心肌细胞凋亡的影响,用18只雄性SD大鼠随机分为Sham组(n=6),TAC组(n=6)和Cartopril组(n=6)。TAC组和Cartopril组大鼠行腹主动脉缩窄术(transverse aortic constriction,TAC)建立高血压模型,Cartopril组术前一周开始给予卡托普利灌胃。术后4周用颈动脉插管法测定各组大鼠血压值,免疫组化法检测GRP78蛋白和CHOP蛋白的表达水平,TUNNEL法检测心肌细胞凋亡。结果①Cartopril组平均动脉压为(124.67±6.26)mmHg,显著低于TAC组(169.73±6.58),但高于Sham组(117.08±7.92),差异有统计学意义(P<0.05);TAC组平均动脉压高于Sham组,差异有统计学意义(P<0.05)。②GRP78在Cartopril组的平均光密度值为0.20±0.02,显著低于TAC组(0.35±0.03),但略高于Sham组(0.16±0.03),差异有统计学意义(P<0.05);TAC组GRP78蛋白表达的平均高密度值高于Sham组,差异有统计学意义(P<0.05)。③CHOP在Cartopril组表达的平均光密度值为0.23±0.03,显著低于TAC组(0.46±0.05),但高于Sham组(0.07±0.02),差异有统计学意义(P<0.05);TAC组CHOP蛋白表达的平均光密度值高于Sham组,差异有统计学意义(P<0.05)。④Cartopril组可见少量凋亡心肌细胞(18.73±0.28)%,显著低于TAC组(31.67±0.54)%,但略高于Sham组(13.49±0.12)%,差异有统计学意义(P<0.05);TAC组细胞凋亡率显著高于Sham组,差异有统计学意义(P<0.05)。说明卡托普利能够通过降低心肌组织内质网应激反应水平,对高血压心肌细胞有保护作用,其作用机制可能与其降低CHOP介导的ERS凋亡反应有关。To study Effects of Cartopril on expression of GRP78 and CHOP in endoplasmic reticulum stress of rat heart after aortic constriction and cardiac myocyte apoptosis,the rats which received abdominal transverse aortic constriction (TAC) were used as experiment models. Carotid artery cannulation was used to measure the blood pressures. Western blot and immunohistochemistry was used to assess the protein level of GRP78 and CHOP. TUNNEL was used to detect the cardiac myocyte apoptosis. All methods were applied to identify:(1) the effect of Cartopril on the expressions of GRP78 and CHOP protein; (2) the effect of Cartopril on cardiac myocyte apoptosis.The results are:(1) The mean arterial pressure of rats in Cartopril group was (124.67 ± 6.26) mmHg,significantly lower than the rats in TAC group [(169.73 ± 6.58) mmHg],but higher than the rats in Sham group[(117.08±7.92) mmHg],the deviation was statistically significant (P0.05). (2) The mean optical density of GRP78 in the rats in Cartopril group was 0.20 ± 0.02,significantly lower than the rats in TAC group (0.35 ± 0.03),but slightly higher than the rats in Sham group (0.16± 0.03),the deviation was statistically significant (P0.05). (3) The mean optical density of CHOP in the rats in Cartopril group was (0.23±0.03),significantly lower than the rats in TAC group (0.46±0.05),but slightly higher than the rats in Sham group (0.07±0.02),the deviation was statistically significant (P0.05). (4) Cartopril group showed a small amount of apoptosis in cardiac myocytes (18.73 ± 0.28)%,significantly lower than the TAC group (31.67 ± 0.54)%,but slightly higher than the Sham group (13.49 ± 0.12)%,the deviation was statistically significant (P0.05); the incidence of cardiac myocyte apoptosis in TAC group was significantly higher than the ones in Sham group,and the deviation was statistically significant (P0.05).
