摘 要:对紫外线灭活的草鱼出血病病毒(GCHV)F9株,在几种鲤科鱼类培养细胞中诱导产生干扰素的能力及影响干扰素产量的各因素进行了研究。纯化的GCHV在紫外线照射5分钟丧失感染性,但获得了在CAB等5种鲤科鱼类培养细胞中高效诱导产生干扰素的能力。干扰素的诱导需要病毒高复数感染细胞。干扰素主要在诱导后14小时内产生。培养上清中的新生牛血清对干扰素的产量有抑制作用,而在pH6.2~7.8范围内对干扰素产量无明显影响。诱导温度和细胞年龄对诱导产量有明显的影响。用紫外线灭活的GCHV在鱼类培养细胞中诱导干扰素时,放线菌素D具有超诱导作用,同种干扰素具有启动效应。本研究为鱼类干扰素的进一步研究及其基因克隆奠定了基础。The ability of ultra violet inactivated grass carp hemorrhagic virus(GCHVuv) to induce interferon in several cyprinoid cell cultures and the factors affecting the interferon yield were investigated.Purified GCHV lost its infectivity in 5 minutes under UV irradiation,but possessed the ability to induce high titer of interferon in five fish cell lines.High interferon induction need high multiplicity of infection.Interferon was mainly produced in the first 14 hours after induction. The interferon yield was lowered by newborn calf serum in the culture medium,but not affected by the changes of pH from pH 6.2 to pH 7.8. The induction temperature and cell age made a notable impact on the inducting yield of interferon.Superinduction by actinomycin D and priming by the same species of interferon were found in the interferon induction with GCHVuv.This work provides basis for the advanced study of fish interferon and the cloning of its genes.
