益气解毒活血方对NZB/W F1狼疮鼠肾炎肾纤维化的干预作用

摘　　要：目的:观察益气解毒活血方对NZB/W F1狼疮鼠肾炎肾纤维化的干预作用,并探讨其可能的作用机制。方法:随机将5月龄狼疮鼠分为空白对照组(空白组)、益气解毒活血小复方治疗组(中药组)、骁悉治疗组(西药组)、益气解毒活血小复方+骁悉治疗组(中西组)。持续干预治疗8周,并分别于实验第0、2、4、6周收集狼疮鼠24 h尿液进行蛋白定量检测,干预结束后观察不同组别狼疮鼠肾脏病理、肾脏羟脯氨酸(Hyp)含量及肾组织转化生长因子-β_1(TGF-β_1)、结缔组织生长因子(CTGF)mRNA的表达情况。结果:不同治疗组均对NZB/W F1狼疮鼠24 h尿蛋白定量、肾脏Hyp含量、肾脏病理、肾组织TGF-β_1与CTGF mRNA表达有改善作用,但各治疗组之间比较无统计学差异;而肾脏病理显示益气解毒活血小复方+骁悉治疗组肾脏病变程度最轻。结论:益气解毒活血方具备降低NZB/W F1狼疮鼠蛋白尿,改善肾纤维化作用,其机制可能与下调致纤维化因子TGF-β_1、CTGF mRNA表达相关。Objective： To investigate the preventive effects of ＂Yiqi Jiedu Huoxue Recipe＂ on nephrofibrosis（ renal fibrosis） of lupus nephritis in NZB/W FI mice and its mechanisms. Methods ： Five-month old NZB/W F1 mice were randomly divided into four groups as placebo group, herb decoction group, cellcept group and herb-cellcept group and were administered with normal saline, ＂Yiqi Jiedu Huoxue Recipe＂, cellcept, and ＂Yiqi Jiedu Huoxue Recipe＂ plus Cellcept respectivelY. Mice were monitored for proteinuria at 0,2,4,6 weeks. Eight weeks later, renal pathology, hydroxyproline, and mRNA expressions of TGF-β1 and CT- GF in kidney were observed. Results： All treatment groups witnessed between the improvement of 24h urine protein, the level of renal hydroxyproline, and the expressions of TGF-β1 and CTGF mRNA, but there was no statistical difference between all treatment groups（ P 〉 0.05 ）. The severity of renal lesions in herb-Cellcept group was the lightest among all the groups. Conelusions： ＂Yiqi Jiedu Huoxue Recipe＂ can decrease 24 h urine protein and ameliorate lupus nephritis of NZB/W F1 mice possibly by down-regulating the expressions of TGF-β1 and CTGF mRNA.
