摘 要:试验研究日粮添加1%α-酮戊二酸(AKG)对多次脂多糖(LPS)刺激断奶仔猪空肠黏膜蛋白、RNA、DNA浓度和抗氧化能力的影响。选用24头健康杜洛克×长白×大白(7.25±0.23)kg断奶仔猪,随机分成3个处理,每个处理8个重复,公、母各半。对照组和LPS组饲喂基础日粮+1%淀粉、AKG组饲喂基础日粮+1%AKG,在试验第10、12、14、16天分别在LPS组和AKG组仔猪的腹膜注射80μg/kgBW脂多糖,对照组注射相应剂量的灭菌生理盐水。第17天清晨屠宰空腹仔猪,刮取空肠黏膜待测。结果表明:多次LPS刺激显著降低断奶仔猪空肠黏膜RNA/DNA、蛋白/DNA、蛋白质浓度及SOD活性(P<0.05),显著升高MDA含量(P<0.05),而日粮中添加AKG能在一定程度上抑制LPS刺激引起的空肠黏膜蛋白合成减少和抗氧化能力降低,维持肠道屏障完整性。This experiment was conducted to study the effects of α-ketoglutaric acid protein synthesis and antioxidative capacity in jejunal mucosa of weaned pigs chronically challenged with lipopolysaccharide. 24 healthy crossbred Duroc × (Landrace × Largewhite) weanling piglets (7.25 ± 0.23 kg BW) were randomly allocated to 3 treatments (control group, LPS control group and AKG group). Each treatment included 8 replicates, 1 piglet per replicate, male and female in half. The control and LPS control group were fed the basal diet supplemented with 1% starch, while the AKG group was fed the basal diet supplemented with 1% α-ketoglutaric acid (AKG). Piglets in the LPS control and AKG group were injected intraperitoneally with 80 μg/kg·BW lipopolysaccharide (LPS) every other day (on d 10, 12, 14 and 16), whereas piglets in the control group were intraperitoneally injected with normal saline at the same dose. On d 17, all pigs were killed under anesthesia, the jejunal mucosa were collected. The resulted indicated that RNA/DNA, Protein/DNA, protein concentration and SOD activity of jejunal mucosa after LPS challenge were decreased (P 0.05), and the MDA concentration was increased (P 0.05), while dietary supplementation with 1%AKG could inhibit the decrease in protein synthesis and antioxidative capacity in jejunal mucosa induced by LPS challenge, AKG had protecting effects on intestinal barrier.
