水稻愈伤组织液体增殖培养的初步研究

摘  要:以MS培养基为基本培养基,采用液体培养的方式,通过调节2,4-D,6-BA及蔗糖浓度,对粳稻南农4007愈伤组织的增殖进行了初步研究。结果表明:2,4-D浓度在0.0~4.0mg/L时对水稻愈伤组织的增殖和生长有一定的影响,6-BA浓度在0.2~3.2mg/L、蔗糖浓度在10~50g/L时对愈伤组织增殖生长差异不显著,但当2,4-D为2.0mg/L、6-BA为0.8mg/L,蔗糖为20g/L时,其愈伤组织不论从生长状态,还是增殖速度上都较好且褐化程度相对较轻。适合粳稻南农4007愈伤组织液体增殖培养的基本配方为MS+NAA0.5mg/L+2,4-D2.0mg/L+水解络蛋白1g/L+蔗糖20g/L+6-BA0.8mg/L。Calli of rice Nannong 4007 were cultured in the MS liquid media with different concentrations of hormones ( 2,4 - D,6 - BA) and sucrose to study their proliferation. The results showed that 2,4 - D has obvious influences on the growth of rice calli within the concentration 0. 0 - 4. 0mg/L in the liquid media. However ,6 - BA has not obvious effects on calli pro- liferation within the concentration 0. 2 - 3. 2mg/L . From rice callus growth point of view ,the sucrose had not obvious influence on rice calli within concentration 10 - 50g/L too. The combination of hormone 2,4 - D2. 0mg/L ,6 - BA0. 8mg/L and sucrose 20g/L had good effect The best suitable medium for rice callus propagation and growth was MS + NAA0. 5 + 2,4 - D2. 0 + 水解络蛋白 1g/L + 蔗糖 20g/L + 6 - BA0. 8mg/L.
