摘  要:目的评价出生后早期低剂量狄氏剂(Dieldrin)暴露对青春期和成年期ICR小鼠黑质纹状体胶质纤维酸性蛋白(GFAP)表达水平的影响。方法仔鼠自出生后(PND)随机分成5个剂量组,即狄氏剂处理组(20、2、0.2μg/kg)、溶剂对照组(DMSO)和生理盐水对照组(NS),于出生后第3天进行腹腔注射染毒,隔天1次,连续6次。分别于青春期和成年期处死一批动物,用Western blot法检测黑质纹状体GFAP的表达情况。结果小鼠出生后早期接触低剂量狄氏剂可引起黑质纹状体GFAP蛋白水平呈剂量依赖性增高,除了0.2μg/kg组和2μg/kg青春期雌鼠组外,其它剂量组均显著升高GFAP蛋白水平,且成年期明显高于青春期。结论出生后早期接触低剂量狄氏剂可引起黑质纹状体GFAP进行性升高。Objective To evaluate the effects of postnatal low dose dieldrin exposure on nigrostriatal glial fibrillary acidic protein (GFAP) in adolescence and adulthood in mice.Methods ICR mice pups were randomly divided into 5 groups and received different doses of dieldrin (20,2,0.2 μg/kg) respectively through intraperitoneal injections (i.p.) every other day on postnatal days(PND)3~13.Control animals received equivalent injections of the dimethylsulfoxide vehicle or normal salin by the same protocol. Western blot was used to evalute nigrostriatal GFAP protein levels on PND36(adolescence) and PND98 (adulthood).Results GFAP protein levels were increased by postnatal low dose dieldrin exposure in a dose-related manner in both sexes,except in the 0.2 μg/kg group and the adolescence female in 2 μg/kg group.There were greater effects observed in the adulthood than in the adolescence.Conclusions These data suggest that developmental exposure to low dose dieldrin could lead to progressive elevations in nigrostriatal GFAP.
