摘  要:目的研究大蒜素与两性霉素B体外联合应用对白色念珠菌菌丝形成的抑制作用,并检测联合应用时细胞毒性的变化。方法采用微量稀释法测定两类药物联合抗白色念珠菌的作用,采用FICI法对联用结果进行评价。用10%小牛血清RPMI1640培养基诱导菌丝形成,检测大蒜素与两性霉素B联合应用对白色念珠菌芽管率和菌丝形成的影响。通过对细胞形态的观察及MTT法检测联合用药后对人宫颈上皮细胞H8毒性的变化。结果两种药物联用对白色念珠菌表现为协同作用,大蒜素能够加强两性霉素B抑制菌丝形成的能力,联合用药后能够显著降低对H8细胞的毒性。结论大蒜素能够提高两性霉素B抗白色念珠菌的作用,是临床上减少两性霉素B用量,减轻其副作用的最佳选择。Objective To explore the combination of allicin and amphotericin B inhibiting effect of filament formation against Candida albicans in vitro,and detect cytotoxicity variation under the combination of the two drugs.Methods The effect of the combination of the two drugs against Candida albicans was determined by the checkerboard micro-dilution method and the fractional inhibitory concentration index(FICI)was calculated.The filament formation was induced in 10% calf serum-1640.The impact of the combination of allicin and amphotericin B on the rate of hyphae and filament formation of Candida albicans was determined.The H8 cytotoxicity variation under the combination of the two drugs was detected through cellular appearance and MTT.Results Drugs combined together acted synergistically against Candida albicans,and allicin could enhance the ability of amphotericin B to inhibit filament formation.The effect of cytotoxicity was reduced remarkably under the combination of the two drugs.Conclusion Allicin largely enhances the anti-Candida albicans activity of amphotericin B and it is a best selection in clinical applications for reducing the dose and adverse effect of amphotericin B.
