摘  要:目的比较左氧氟沙星、加替沙星和莫西沙星的抗菌活性及对角膜上皮细胞的毒性作用。方法细菌与细胞共孵育1h后,抗生素处理1h,行荧光染色和CFU计数;MTT法和划痕法检测细胞的增生和迁移能力,评价3种抗生素的细胞毒性作用。结果 3种抗生素的抗菌效果均非常显著,其中莫西沙星对金黄色葡萄球菌和左氧氟沙星对绿脓杆菌的抑菌活性最强,对凝固酶阴性葡萄球菌和黏质沙雷菌的抗菌效果差异无统计学意义;在质量浓度低于125mg/L时,左氧氟沙星的毒性明显低于莫西沙星和加替沙星。结论莫西沙星、左氧氟沙星和加替沙星3种抗生素在抗菌活性和上皮细胞毒性方面存在一定差异。BackgroundFuoroquinolones have been widely used in bacterial keratitis because of its rapid bactericidal action.But there is no scientific comparison and evaluation about its antibacterial activity and cellular toxicity on corneal epithelium.ObjectiveThe present study was to compare the bacterial susceptibility and cellular toxicity of levofloxacin,moxifloxacin and gatifloxacin in human corneal epithelial cells(CECs).MethodsThe human corneal epithelial cells strain,ATCC CRL-11135,were incubated together with cultured staphylococcus aureus,coagulase-negative staphylococci,pseudomonas aeruginosa and serratia marcescens in agar plate for 1 hour and then exposed to 1000.000,500.000,250.000,125.000,62.500,31.250,15.625,0mg/L of antibiotics for another 1 hour,separately.The adhesiveness test of bacterium on CECs was assessed by Hoechst33342/EB florescent staining under the confocal microscope,and CFU counting was performed to evaluate the growth of bacterium.MTT assay was used to test the proliferative ability of CECs after usage of different antibiotics,and the migration rate of CECs was detected by wound healing assay to evaluate the toxicity of drugs to cells.The antibiotical activity and drug toxicity were compared among these three drugs.ResultsThe CECs adhesive bacterium showed the red fluorescence for Hoechst33342/EB.The positive cells for Hoechst33342/EB staining in all of the three drug groups were significantly decreased in comparison with control group.The strongest effect was found in moxifloxacin group for antibiotics against staphylococcus aureus and levofloxacin for antibiotics against pseudomonas aeruginosa.However,all these three antibiotics showed the similar efficacy against coagulase-negative staphylococci and serratia marcescens.The migration rates of cells were significantly declined in all of the three antibiotics groups in comparison with control group at a concentration-dependent manner at the range from 62.5mg/L to 500mg/L.Moxifloxacin,levofloxacin and gatifloxacin inhibited the proliferat
