银杏内酯B对CIA大鼠成纤维样滑膜细胞增殖的影响

摘　　要：①目的探讨银杏内酯B对胶原诱导性关节炎（collagen-induced arthritis,CIA）大鼠成纤维样滑膜细胞（Fibroblast-like synovi-al cells,FLS）的影响。②方法雄性Wistar大鼠皮内注射Ⅱ型胶原,建立CIA大鼠模型,然后取其滑膜组织用胶原酶消化法进行原代培养,取3~5代的滑膜细胞。应用MTT比色法检测1×10-9、1×10-8、1×10-7、1×10-6、1×10-5mol/L的银杏内酯B作用24、48和72h后FLS的增殖水平。③结果（1×10-9~1×10-5）mol/L的银杏内酯B作用24、48和72h,均能明显抑制FLS的增殖（P〈0.05）,且具有时间、剂量依赖性。④结论银杏内酯B能明显抑制CIA大鼠FLS的增殖并呈时间、剂量依赖性改变。Objective To investigate the effect of the ginkgolide B on fibroblast-like synovial cells（FLS）of rat with collagen-induced arthritis（CIA）.Methods The model of CIA in male wistar rats was established by intradermal injecting the typeⅡcollagen,then the synovial tissue was taken and digested by collagenase for primary culture,and 3 to 5 generations of synovial cells were used.After fibroblast-like synoviocytess were treated with ginkgolide B（1×10-9,1×10-8,1×10-7,1×10-6,1×10-5mol/L） for 24,48 and 72h,the proliferation of fibroblast-like synoviocyte was examined using MTT colorimetric assay.Results The proliferation of FLS was inhibited by 1×10-9~1×10-5mol/Lginkgolide B at 24,48 and 72h（P0.05） and had time and dose-dependent.Conclusion The proliferation of FLS in rat with CIA could inhibited by ginkgolide B and was time dose-dependent change.
