干细胞移植治疗兔下肢肢体缺血的实验研究

摘　　要：目的探讨体外培养的自体骨髓间充质干细胞治疗兔下肢肢体缺血的治疗效果.方法将体外培养扩增后获得的自体骨髓干细胞悬液分别注射于家兔右下肢缺血模型的股动脉周围的大腿肌肉群内.术后3、6周后通过血管造影术及病理学方法,检测患肢侧支循环和血管新生情况.结果体外培养的自体骨髓间充质干细胞治疗兔下肢肢体缺血取得了满意的疗效.结论体外培养的自体骨髓间充质干细胞不能治疗下肢缺血性疾病的一种有效方法.Objective To investigate the therapeutic effect of transplanting the autologous bone marrow mesenchymal stem cells cultured in vitro in the treatment of lower limbs ischemia in rabbits.Method The suspension of autologous bone marrowmesenchymal stem cells was injected into the muscles of schemic lower limbs models,then 3 and 6 months later,the collateral circulation and blood vessel neogenesis was examined by DSA and pathological approaches.Result The autologous bone marrow mesenchymal stem cells got satisfactory therapeutic effects in the treatment of lower limbs ischemia in rabbits.Conclusion The transplantation of autologous bone marrowmesenchymal stem cells cultured in vitro is an effective therapy in the treatment of ischemic disease of lower limbs.
