不同剂量PPDA接触致敏中Th1/Th2的调节作用

摘  要:目的研究不同剂量的PPDA诱导免疫耐受的反应曲线和免疫耐受过程中Th1细胞和Th2细胞的调节关系。方法用不同剂量的PPDA建立3个不同的动物实验模型,连续12周,用临床皮肤评分法得到这3个实验模型免疫反应的曲线,并比较其强弱;在实验的4个阶段取其活组织用ELISA法进行IL-10,IL-12,IL-1α细胞因子测定,分析IL-10和IL-12在免疫耐受发生中的调节关系。结果3组别各周平均反应分值无差别,在第7和9周实验剂量交叉时,大剂量组的皮肤反应大于小剂量组。IL-10皮肤含量在实验第6周达高峰,a1和a2组的IL-10含量在第8周较第6周明显下降,差异有统计学意义(P<0.05);a1组的IL-10值在第12周较第6周明显下降,差异有统计学意义(P<0.05);a1组在第12周的IL-10含量低于a2组。第6周的IL-12含量大于0时刻,差异有统计学意义(P<0.05);第8周IL-12含量较第6周明显上升;a1组在第12周的IL-12大于a2组。IL-1α各组各周之间差异均无统计学意义(P>0.05)。结论在本试验条件下,小剂量抗原较大剂量抗原更易使Th1向Th2倾斜;IL-10可能调节Th2型免疫反应。Objective To find the response curves induced by different concentrations of PPDA and the relationship between Th1 and Th2 in regulating the Th1 type immune reaction. Methods Establishing three different animal test models with different concentrations of PPDA. The test period was 12 weeks. Biopsies were taken four times during the whole test. The IL-10,IL-12 and IL-1α contents were quantified and the relationship between Th1 and Th2 was analysed finally. Results The results showed that the scales of skin reaction were not different significantly ( P 0. 05) among three groups during the whole test. The scales of skin reaction of higher concentration group were stronger than those of the lower one at W7 and W9. The level of IL-10 was peaked at W6 in these 3 groups. The level of IL-10 of group 1 and 2 at W8 decreased compared to W6 and the difference was statistically significant (P 0. 05). The level of IL-10 of group 1 at W12 decreased compared to that at W6 and the difference was significant (P 0. 05). The level of IL-10 of group 1 was lower than that of group 2 at W12. The level of IL- 12 at W6 was higher than that at W0 and the difference was significant ( P 0. 05). The level of IL-12 at W8 was higher than that at W6. The level of IL-12 of group 1 was higher than that of group 2 at W12. The level of IL-1α was not significantly different (P 0. 05) among three groups at the same time point of the whole test. Conclusions These findings indicated that Th1 was easier switch to Th2 induced by higher-concent allergen than lower one. Further more,IL-10 could be a early regulator of Th2 type immune reaction.
