摘  要:检测上海地区就医人群血液中针对抗生素类药物抗药物抗体的产生情况。选取在上海市血液中心进行交叉配血与Coombs'试验及进行骨髓移植的患者血浆,选取上海地区使用率靠前的9种抗生素类药物包被的RBC,采用试管法与凝胶法进行间接抗球蛋白实验,检测血浆中是否有抗药物抗体的存在。结果成功制备了青霉素包被的RBC并得到证实;所有被检测的181个样本中,有8例样本(4.42%)发现有抗青霉素特异性抗体的存在;1例样本(0.55%)存在抗头孢菌素类药物的抗体;还有2例样本(1.1%)含有非特异性抗药物抗体。在适当的缓冲体系中及一定浓度的药物溶液中可以成功制备药物包被的RBC并用于血浆中抗药物抗体的检测。有一定比例的就医人群在使用某些抗生素后会产生抗药物抗体,可以通过使用药物包被的RBC得到检测。To detect production of anti-antibiotics antibodies in the blood samples of population under medical care in Shanghai, blood plasma samples were taken from patients whose blood samples were undergone examination for blood cross-matching, Coomb's test or bone marrow transplantation in Blood Bank Center of Shanghai,and the indirect antiglobulin test(IAT) by using 9 kinds of top-ranking antibiotics-coated RBCs was used to detect the presence of antibodies against antibiotics in plasma. In these ways,the penicillin-coated RBCs were prepared successfully and the anti-penicillin specific antibodies were demonstrated in 8 plasma samples(4.42%).Meanwhile,antibodies to cephalosporins existed only in one case of samples(0.55%);and there were two cases of samples(1.1%) with non-specific anti-drug antibodies in all the tested 181 samples.It is evident that the drug-coated RBCs can be prepared successfully in appropriate buffer system and certain concentration of the drug solution and it can be used for the detection of anti-drug antibodies in plasma.The production of certain anti-drug antibodies can be confirmed by using the drug-coated RBCs in certain percentage of patients under medical care.
