摘 要:【目的】观察骨髓基质干细胞(BMSCs)种植到复合胶原和生长因子的聚乙醇酸-乳酸共聚物(PLGA)生物材料上,再种植到兔体内,进一步诱导成组织工程软骨组织的可行性,并观察此组织工程软骨修复兔关节软骨缺损的能力。【方法】相分离法制作PLGA,复合Ⅱ型胶原和成软骨生长因子bFGF,TGF-β1。将P3代的BMSCs种植到复合材料上。36只新西兰大白兔随机分为实验组、对照组、空白组,分别于肌袋内植入BMSCs/复合生长因子和胶原的PLGA、复合生长因子和胶原的PLGA、复合胶原的PLGA,于术后第4、8、12周取材观察细胞的定向分化及生长情况,包括大体观察、苏木精-伊红染色、甲苯胺蓝染色、Ⅱ型胶原染色、扫描电镜观察。16只新西兰白兔分2组,上述组织工程骨植入实验兔膝关节软骨缺损处,对照组仅做缺损,12周后取材初步观察修复情况。【结果】大体观察可见实验组材料有类软骨样组织生长,而对照组和空白组则仅见纤维组织生长。各种染色及电镜观察显示:实验组复合物内可见多的成软骨细胞及少量的破骨细胞。实验组甲苯胺蓝染色和Ⅱ型胶原染色为阳性,对照组和空白组均为阴性。大体观察兔膝关节软骨缺损得到修复,苏木精-伊红染色显示表面关节软骨形成,与周围正常软骨形成连接。【结论】胶原修饰的PLGA生物材料具有较好的细胞相容性;BMSCs种植到复合胶原和生长因子的PLGA生物材料上在兔体内可诱导成为组织工程软骨复合组织,并初步证实有修复兔关节软骨缺损的能力。【Objective】To observe the condition of constructing the tissue engineered osteochondral composite through bone marrow stem cells(BMSCs)cultured on the poly-lactide-co-glycolic acid(PLGA),which is modified with collagen and cellular growth factors.In order to explore a new method of constructing the tissue engineered chondral composite.Then observe the effect of the composite cartilage on repairing of articular cartilage defect to rabbits.【Methods】PLGA was made by phase separation technique,composite with collagen typeⅡ,basic fibroblast growth factor and transforming growth factor-β1.The third era(P3) BMSCs were cultured on the above scaffold.Thirty-six New Zealand white rabbits were divided into experimental group,control group,blank group at random.These groups were implanted with BMSCs/growth factors and collagen-PLGA,growth factors and collagen-PLGA and collagen-PLGA into the muscle respectively.Then observe the directional differentiation and growth condition ofcells at the 4th,8th,and 12th week after the operations.Including general shape observation,HE,toluidine blue,collagen typeⅡ staining and the cells shape observation through scanning electron microscope.Sixteen New Zealand white rabbits were divided into two groups at random.The composite materials were planted into the cartilage defects of the keen of the experimental group,while the control group was left untreated.Then observe the effect of the repairing condition at the 12th week after the operation.【Results】There was cartilage-like tissue growing in experimental group in its general shape,while there was only fibrous tissue growing in control group and blank group.Various staining and scanning electron microscope observation showed:there were many cartilages and some osteoblasts in the composite of experimental group.The toluidine blue and collagen typeⅡstaining in experimental group were both positive,while in control group and blank group were negative.In the defect experiment,the defects were repaired,and the HE
