福莫特罗对哮喘小鼠气道杯状细胞增生及粘蛋白MUC5ac mRNA表达的影响

摘　　要：【目的】探讨长效β2受体激动剂福莫特罗对哮喘(支气管哮喘)小鼠气道杯状细胞增生及粘蛋白MUC5acmRNA表达的影响。【方法】40只雌性SPF级BABL/c小鼠,随机分为4组,每组10只。A组为生理盐水对照组;B组为卵蛋白(ovalbumin,OVA)哮喘组;C组为哮喘福莫特罗干预组;D组为哮喘地塞米松干预组。在末次激发24h后所有小鼠取左肺组织行过碘酸雪夫(PAS)染色,采用医学图像分析软件测定支气管上皮杯状细胞面积占上皮细胞总面积的百分比并计算杯状细胞数量。取右肺组织行real-timeqRT-PCR,检测粘蛋白MUC5acmRNA的表达。【结果】B组小鼠支气管上皮杯状细胞的数量、杯状细胞占上皮细胞总面积的百分比和MUC5acmRNA水平显著高于A组[(163.63±16.68)个vs(0.46±0.16)个,(77.36±5.05)%vs(0.03±0.01)%,(10.31±0.73)vs(1.00±0.13),P均<0.05];C组小鼠支气管上皮杯状细胞的数量、杯状细胞占上皮细胞总面积的百分比和MUC5acmRNA水平均低于B组[(52.04±4.60)个vs(163.63±16.68)个,(30.05±3.72)%vs(77.36±5.05)%,(1.64±0.14)vs(10.31±0.73),P均<0.05];D组小鼠支气管上皮杯状细胞的数量、杯状细胞占上皮细胞总面积的百分比和MUC5acmRNA水平均低于B组[(63.41±6.39)个vs(163.63±16.68)个,(38.52±3.83)%vs(77.36±5.05)%,(1.72±0.10)vs(10.31±0.73),P均<0.05];C组和D组的杯状细胞数量、面积百分比和MUC5acmRNA水平间差异均无统计学意义[(52.04±4.60)个vs(63.41±6.39)个,(30.05±3.72)%vs(38.52±3.83)%,(1.64±0.14)vs(1.72±0.10),P均>0.05]。【结论】长效β2受体激动剂福莫特罗可抑制哮喘小鼠气道杯状细胞增生及粘蛋白MUC5acmRNA的表达。【Objective】To investigate the effect of long-acting β2-adrenoceptor agonist formoterol on airway goblet cell hyperplasia and protein MUC5ac mRNA expression in asthmatic mice.【Methods】Forty female BABL/c mice were randomly divided into four groups with 10 mice in each.Mice in group A were treated with saline as control,and mice in group B,group C,and group D were sensitized by OVA to establish asthmatic model,but group C were pretreated with formoterol and group D were pretreated with dexamethasone.All mice were killed 24 hours after the final OVA challenged.The left lung tissue sections were stained with periodic acid Schiff（PAS）for identification of goblet cell hyperplasia.The right lung was isolated for detecting MUC5ac mRNA by the method of real-time fluorescence quantitative reverse transcription polymerase chain reaction（real-time qRT-PCR）.【Results】The number of the goblet cell,the percentage of goblet cell to total cell and the expression of MUC5ac mRNA in group B were significant higher than those of group A[（163.63±16.68）vs（0.46±0.16）,（77.36±5.05）%vs（0.03± 0.01）%,（10.31±0.73）vs（1.00±0.13）,P0.05].The number of the goblet cell,the percentage of goblet cell to total celland the expression of MUC5ac mRNA in group C were significant lower than those of group B[（52.04±4.60）vs（163.63±16.68）,（30.05±3.72）%vs（77.36±5.05）%,（1.64±0.14）vs（10.31±0.73）,P0.05].The number of the goblet cell,the percentage of goblet cell to total cell and the expression of MUC5ac mRNA in group D were significant lower than those of group B[（63.41± 6.39）vs（163.63±16.68）,（38.52±3.83）%vs（77.36±5.05）%,（1.72±0.10）vs（10.31±0.73）,P0.05].For the index mentioned above,there were no significant differences between group C and group D[（52.04±4.60）vs（63.41±6.39）,（30.05±3.72）%vs（38.52±3.83）%,（1.64±0.14）vs（1.72±0.10）,P0.05].【Conclusions】This study demonstrated that the long-actingβ2-agonist formoterol could in
