摘 要:目的探讨一氧化氮(NO)、鸟苷酸环化酶(GC)在扩张型心肌病小鼠心肌内的表达及其与心肌损害的关系。方法将4~5周龄Balb/c小鼠随机分成实验组和正常对照组;以柯萨奇病毒B3(CVB3)感染Balb/c小鼠建立扩张型心肌病小鼠模型;通过比色法检测NO的变化,RT-PCR法测定外周血GC mRNA,HE、VG染色观察心肌损害程度。结果①小鼠感染3个月后心脏收缩功能明显减退[LVEF(61.74±7.47)%比(91.59±1.38)%];6个月后心室壁活动度明显减弱,左室收缩功能进一步减退[6个月LVEF为(50.38±12.91)%),9个月(46.57±8.32)%],与3、1个月时比较,差异有统计学意义(P<0.05)。②实验组NO水平1个月(40.48±9.89)μmol/L、3个月(46.37±11.23)μmol/L、6个月(74.57±15.26)μmol/L逐渐升高,9个月(32.54±7.63)μmol/L有所下降,但与对照组(25.09±5.54)μmol/L比较差异均有统计学意义(P<0.01)。③实验组GC mRNA表达于CVB3感染1个月(0.575±0.081)、3个月(0.763±0.056)、6个月(0.835±0.044)逐渐升高,与对照组(0.441±0.028)比较差异有统计学意义(P<0.01),9个月(0.482±0.037)虽有所下降,但与对照组比较差异无统计学意义(P>0.05)。④实验组NO表达与心肌病理积分呈显著正相关(r=0.548,P<0.05),GC mRNA表达量与NO亦呈显著正相关(r=0.613,P<0.05)。结论扩张型心肌病小鼠的心肌内表达了NO、GC,与心肌损害成正比。Objective To examine the myocardial expressions of NO and guanylate cyclase(GC) in mice with dilated cardiomyopathy,and analyze the relationship between their expressions and myocardial damage.Methods One hundred 4-5 weeks Balb/c mice were randomly divided into two groups: normal control group and experiment group.Dilated cardiomyopathy model was established by subjecting Balb/c mice to Coxsackie virus B3(CVB3) infection.The heart tissues were examined at 1,3,6,9 months after CVB3 infection.Expression of NO was measured by colorimetry methods,GC mRNA was determined by RT-PCR,myocardial damage was assessed by HE and VG staining.Results(1) Left ventricular systolic function was significantly decreased at 3 months in experimental group(LVEF 61.74% ± 7.47% vs 91.59% ± 1.38% in controls).Ventricular wall activity was significantly decreased,and left ventricular systolic function was further impaired at 6 and 9 months,the differences were significant compared with 1 month and 3 months time points(P〈0.05).(2) The expressions of NO in experiment group were 40.48 ± 9.89 μmol/L,46.37 ± 11.23 μmol/L,74.57 ± 15.26 μmol/L,and 32.54 ± 7.63 μmol/L,respectively,at 1,3,6,9 months,the difference between two groups at each time point was significant(P〈0.01).(3) The expressions of GC mRNA in experiment group were 0.575 ± 0.081,0.763 ± 0.056,0.835 ± 0.044,and 0.482 ± 0.037,respectively,at 1,3,6,9 months.The difference between two groups was significant at 1,3,6 months,and without significance at 9 months.(4) NO expression in the experiment group was significantly correlated with cardiac pathology score(r = 0.548,P〈0.05).GC mRNA expression in experiment group significantly correlated with NO expression(r = 0.613,P〈0.05).Conclusions Expressions of NO and GC were found in mice with dilated cardiomyopathy,and they were positively associated with the severity of myocardial injury.
