摘 要:目的研究新生期大鼠反复惊厥后皮层中微管相关蛋白1轻链3(LC-3)的表达及3甲基腺嘌呤(3-MA)对表达的调控作用。方法生后6 d的SD大鼠随机分成3组,每组24只,惊厥组每日吸入三氟乙醚诱导惊厥发作5次,每次间隔30 min,连续9 d;对照组同样操作但不吸入三氟乙醚;3-MA组惊厥前腹腔注射3-MA(总量2μl),同样方法吸入三氟乙醚诱导惊厥,方法同惊厥组。3组大鼠于最后一次惊厥后分别按1.5、3、6、24 h后断头取脑,采用蛋白质免疫印迹技术(Western blot)分别检测3组大鼠大脑皮层中LC-3的表达。结果惊厥组(1.5、3、6 h)LC-3的表达明显高于对照组(t=2.483,2.599,6.937,1.280均P<0.05),3-MA组LC-3的表达明显低于惊厥组(t=2.243,7.775,5.887,4.245均P<0.05)。而24 h惊厥组LC-3的表达并不明显高于对照组(t=1.280,P>0.05)。结论惊厥后自噬途径被激活;3-MA参与了自噬途径的调控。Objective To investigate dynamic expression of cortex microtubule-associated protein 1 light chain 3(LC-3) and intervention effect of autophagy/lysosome inhibitor 3-methlyadenine(3-MA) in neonatal rat with recurrent seizures.Methods Seventy-two 6-day-old SD rats were randomly divided into three groups(24 rats in each): recurrent seizures group(RS group),3-MA-treated group(3-MA group),and control group.Rats in RS group were induced to seizures 5 times each day for 9 days with aspiration of flurothyl during the first 12 days of life.Rats in 3-MA group were induced to seizures with the same method,and were treated with intraperitoneal injection of 3-MA before seizure-induction each day(totally 2 μl).The brains of the rats were taken at 1.5,3,6,and 24 h after the last convulsion.Western blot was employed to determine alternations in LC3 in cortex at different time points.Results LC-3 expression was significantly higher in RS group than that of the control group at 1.5,3,and 6 h(P〈0.05),while the difference between two groups at 24 hours after seizure was not significant.LC-3 expression decreased significantly in 3-MA group compared with those in RS group at all time points(P〈0.01).Conclusions Autophagy/lysosomal pathway were activated immediately after recurrent seizures as indicated by the elevated expression of LC3 in cortex.3-MA was involved in the regulation of autophagy/lysosomal pathway.
