摘  要:目的探讨促肾上腺皮质激素[ACTH(1-24)]对脐静脉内皮细胞株(ECV304)和星形胶质细胞的细胞毒性作用。方法采用体外细胞培养技术,以细胞形态学、细胞活性、乳酸脱氢酶(LDH)释放量及细胞凋亡率的改变为指标,观察不同浓度的ACTH(1-24)对ECV304和星形胶质细胞的影响。结果 ACTH(1-24)血药浓度在1μg/ml、5μg/ml、25μg/ml、100μg/ml、200μg/ml范围内对ECV304和星形胶质细胞的形态、细胞活性、凋亡率及LDH的释放均无明显影响,组间比较差异无统计学意义(P>0.05)。结论 ACTH(1-24)在1~200μg/ml的浓度范围内对ECV304和星形胶质细胞均无毒性作用。Objective To study the cytotoxic effect of ACTH(1-24)to umbilical vein endothelial cell strain(ECV304)and astrocyte.Methods The technology of invitro cell culture was adopted.Cytomorphology,cellular activity,release of actic dehydrogenase(LDH)and the change of apoptosis rate were analyzed as the indicators to observe the effects of different ACTH(1-24)concentrations on ECV304 and astrocyte.Results The blood concentrations of ACTH(1-24)at 1 μg/ml,5 μg/ml,25 μg/ml,100 μg/ml and 200 μg/ml had no significant effect on cytomorphology,cellular activity,the release of LDH,the change of apoptosis rate of ECV304 and astrocyte with no significant difference when compared between the groups(P0.05).Conclusion ACTH(1-24)concentrations in the range of(1~200 μg/ml)has no cytotoxic effect on ECV304 and astrocyte.
