摘 要:目的研究卡托普利、缬沙坦及中药银杏叶提取物(Ginkgo bilobaextract,GBE)对高糖培养的大鼠肾小球系膜细胞(MC)的影响,探讨三者单独应用及中西药联合应用时的异同,从而为GBE与卡托普利或缬沙坦联合应用提供理论基础。方法体外培养HBZY-1大鼠MC,分为9组(每组n=3):①正常对照(NC)组;②甘露醇(MA)组;③高糖(HG)组;④溶媒(DMSO)组;⑤GBE组;⑥卡托普利(Cap)组;⑦缬沙坦(Val)组;⑧GBE和卡托普利联合用药(GC)组;⑨GBE和缬沙坦联合用药(GV)组。可见光分光光度法测定胞质超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、总抗氧化能力(T-AOC)、谷胱甘肽过氧化物酶(GSH-Px)抗氧化指标的水平,RT-PCR法测定细胞转化生长因子β1(TGF-β1)mRNA的相对含量。结果GBE、卡托普利、缬沙坦以及联合用药均可提高高糖环境中MC的抗氧化能力,且联合用药组的作用更强;GBE、卡托普利、缬沙坦以及联合用药均可抑制高糖诱导的TGF-β1mRNA水平上调,联合用药组作用较强。结论中西药联合应用时对大鼠MC的细胞保护作用优于单独用药。Objective To investigate the effect of captopril,valsartan and Chinese crude drug Ginkgo biloba extract(GBE) on rat glomerular mesangial cells cultured in high glucose and to probe into the differences between monotherapy and combined treatment with the drugs so as to provide rationale for combination of GBE with captopril or valsartan.Methods Rat glomerular mesangial cell line(MC) HBZY-1 was cultured in vitro in DMEM media.The cells were divided into 9 groups(n=3 each): ① normal control(NC) group;② mannitol(MA) group;③ high glucose(HG) group;④ solvent control(DMSO) group;⑤ GBE group;⑥ captopril(Cap) group;⑦ valsartan(Val) group;⑧ captopril and GBE combination therapy(GC) group;⑨ valsartan and GBE combination therapy(GV) group.Following each corresponding treatment,superoxide dismutase(SOD),catalase(CAT),total antioxygen capability(T-AOC) and glutathione peroxidase(GSH-Px) in MC were assayed by visible spectrophotometry;transforming growth factor-β1(TGF-β1) mRNA was determined by RT-PCR.Results Monotherapy or combination therapy with GBE,captopril and valsartan can promote the antioxygen ability,while combination therapy had better effect than monotherapy.Treatment with GBE,captopril and valsartan either individually or in combination could inhibit TGF-β1 mRNA up-regulation induced by high glucose,while combination therapy had stronger effects.Conclusions Combination therapy with Chinese crude drugs and western medicine is superior to monotherapy in protection of rat glomerular mesangial cells.
