摘  要:目的探讨γ-分泌酶抑制剂DAPT对人宫颈癌HeLa细胞体外增殖的抑制作用及相关机制。方法MTT法检测不同浓度DAPT对HeLa细胞的增殖抑制作用;RT-PCR法检测HeLa细胞Notch1、HES1基因表达情况。结果不同浓度的DAPT作用于HeLa细胞后,细胞增殖水平明显下降(P<0.05或P<0.01),且DAPT对HeLa细胞增殖的抑制作用呈剂量依赖性。随着DAPT浓度的增加,Notch1、HES1 mRNA的表达逐渐下调。结论DAPT能够抑制HeLa细胞的增殖,可能与HeLa细胞中Notch1、HES1 mRNA的下调有关。Objective To explore the effects of γ-secretase inhibitor DAPT on the proliferation in vitro of human cervical carcinoma cell line HeLa and its underlying mechanism.Methods The inhibitive effects of DAPT at different concentration on the proliferation of HeLa cells was detected by MTT assay;the expressions of Notch1 and HES1 mRNA were evaluated by reverse transcrip tase-polymerase chain reaction(RT-PCR) analysis.Results Following treatment with DAPT at different concentrations,the growth and proliferation of human HeLa cells significantly decreased(P0.05 or P0.01),DAPT produced antiproliferative effects on HeLa cells in a dose-dependent manner.The mRNA expression of Notch1 and HES1 was gradually downregulated with the increase of DAPT doses.Conclusion The γ-secretase inhibitor DAPT can inhibit the proliferation of HeLa cells,which might be related with the down-regulation of the mRNA expression of Notch1 and HES1.
