摘 要:目的观察肠易激综合征(Irritable bowel syndrome,IBS)模型大鼠远端结肠组织5-羟色胺(5-hydroxytryp-tamine,5-HT)和5-羟吲哚乙酸(5-hydroxyindoleacetic acid,5-HIAA)的表达以及眼针干预的影响。方法实验动物分成正常组、模型组、眼针组,采用束缚刺激与应激刺激相结合的方法建立IBS动物模型并进行眼针干预。各组采用ELISA法检测血清和远端结肠组织中5-HT和5-HIAA水平。结果与正常组比较,模型组5-HT和5-HIAA水平均显著升高,5-HT转化率也显著升高,但5-HT的增加量明显高于5-HIAA的增加量,提示5-HT降解与合成分泌的不相匹配可能是模型组大鼠5-HT水平升高的重要原因;而与模型组比较,眼针组5-HT和5-HIAA水平明显下降,眼针组5-HT转化率显著降低。结论 5-HT降解与合成分泌的不相匹配可能是模型组大鼠5-HT水平升高的重要原因,眼针干预可有效改善IBS大鼠5-HT代谢,使5-HT恢复至正常水平。Objective To investigate the expressions of 5-hydroxytryptamine (5-HT) and 5-hydroxyindoleacetic acid (5-HIAA) in distal colonic tissues and the effect of eye acupuncture intervention in a rat model of irritable bowel syndrome (IBS). Methods Experimental animals were allocated to normal,model and eye acupuncture groups. An animal model of IBS was made by combined binding and stress stimulation. Eye acupuncture intervention was given. 5-HT and 5-HIAA levels in serum and distal colonic tissues were measured by ELISA in each group. Results 5-HT and 5-HIAA levels increased obviously and 5-HT transformation rate also did in the model group compared with the control group. But an increase in 5-HT was significantly higher than that in 5-HIAA,indicating that the inconsistency between 5-HT degradation and synthesis-secretion might be the important cause of increase in 5-HT levels in the model group of rats. 5-HT and 5-HIAA levels decreased significantly and 5-HT transformation rate also did in the eye acupuncture group compared with the model group. Conclusion The inconsistency between 5-HT degradation and synthesis-secretion may be the important cause of increase in 5-HT levels in the model group of rats. Eye acupuncture intervention can effectively improve 5-HT metabolism and restore 5-HT to a normal level in IBS rats.
