摘 要:[目的]探索双酚A(BisphenolA,BPA)对断乳期F1雄性子代SD大鼠生殖和发育的影响。[方法]从孕0d对母鼠染毒50mg/kg、100mg/kg、200mg/kg BPA至断乳期停止染毒,对睾丸的Bax、Bcl-2、PCNA、ER的表达进行免疫组织化学分析。[结果]图象分析结果表明,随着BPA染毒剂量的增高,Bax表达的细胞灰度值有降低的趋势,50mg/kg、100mg/kg和200mg/kg BPA组的Bax与对照组相比,差异均有统计学意义(P﹤0.05);而Bcl-2表达的细胞灰度值有增高的趋势;100mg/kg、200mg/kg BPA组的Bcl-2与对照组相比,差异有统计学意义(P﹤0.05);PCNA表达的细胞灰度值50mg/kg,100mg/kg,200mg/kg BPA组与对照组相比,差异均有统计学意义(P﹤0.05);生精细胞ER表达的灰度值有增高的趋势,50mg/kg,100mg/kg,200mg/kg BPA组与对照组相比,差异均有统计学意义(P﹤0.05)。[结论]BPA导致F1雄性子代生殖发育毒性的机制,可能是导致生精细胞凋亡增强,增殖减慢,细胞内转录、表达受阻。[Objective] To study the effect of BPA on the development of reproductive system of F1 generation male rats during weaning period by tests.[Methods] The female rats were exposed to bisphenol-A from 0 day after pregnancy to the weaning period at doses of 50,100 and 200 mg /kg,respectively.Immune histochemical analysis was also carried out to investigate the expression of Bax,Bc1-2,PCNA,and ER of testis.[Results] Immune histological analysis showed that the expression of Bax in the epithlia of testis seminiferous tubules increased while the expression of Bcl-2 and PCNA decreased in three experiment groups indicating a lagged development in seminiferous tubules.There was significant difference among the expression of Bax of 50 mg /kg,100 mg /kg and 200 mg /kg BPA group and the control group(P﹤0.05);When compared the expression of Bcl-2 of 100 mg /kg and 200 mg /kgBPA groups with the control group,there were statistically significant(P﹤0.05);When compare the PCNA expression of 50 mg /kg,100 mg /kg and 200 mg /kgBPA groups with the control group,there were statistically significant(P﹤0.05);Rats in experimental groups had weaker expression of ER of epithlia of testis seminiferous tubules than those in the control;When compare the expression of ER of 50 mg /kg,100 mg /kg and 200 mg /kg the BPA groups with the control group,there were statistically significant(P﹤0.05).[Conclusion] This study showed that the possible mechanism of obvious reproductive and developmental toxicity in F1 male rats by BPA is typical estrogen effect,and influence the normal endocrine function,which may increase apoptosis,decrease proliferation and inhibite the transmission and expression of spermatogenic cells.
