摘  要:目的探讨组织蛋白酶S(CatS)、组织蛋白酶K(CatK)和胱抑素C在压力负荷性心肌肥厚左心室重构过程中的表达及其相互关系。方法雄性Wistar大鼠60只被随机分配到4周假手术组(F4)、8周假手术组(F8)、4周手术组(T4)以及8周手术组(T8)。采用腹主动脉缩窄法构建心肌肥厚模型和心力衰竭模型。用实时荧光定量PCR和免疫组化法分别检测CatS、CatK和胱抑素CmRNA及蛋白表达,用Masson染色法分析左心室肌间质和血管周围纤维化程度,心脏超声和血流动力学测定心功能和心室重构。结果实时荧光定量聚合酶链式反应(PCR)结果显示T4组大鼠心肌中的CatS、CatK及胱抑素CmRNA的表达均明显高于F4组大鼠(均P<0.05)。T8组大鼠心肌中的CatS、CatKmRNA的表达进一步升高,显著高于F8组(P<0.01),而T8组大鼠心肌中的胱抑素CmRNA的表达则降至正常水平。增高的CatS、CatK及胱抑素C蛋白主要表达于心肌细胞。结论 CatS、CatK和胱抑素C参与了压力负荷性心肌肥厚左心室的重构。Objective To observe the expression of cathepsin S,K and cystatin C during the left ventricular remodeling in rats with pressure overloaded myocardial hypertrophy.Methods Sixty male wistar rats were randomized into 4-week sham-operated group(F4),8-week sham-operated group(F8),4-week aortic-banded group(T4)and 8-week aortic-banded group(T8).Hypertrophy model(H-LVH)and Heart Failure model(H-HF)was established by abdominal aortic constriction.The expression of cathepsin S,K and cystatin C was detected by real-time quantitative PCR and by immunohistochemical method.The interstitial fibrosis and perivascular fibrosis was analyzed with Masson's staining.Cardiac function was evaluated by echocardiographic and hemodynamic technique.Results Real-time quantitative PCR analysis revealed that the expression of Cat S,Cat K and cystatin C mRNA was increased significantlyin T4 group compared with F4 group.The expression of Cat S,Cat K mRNA was increased dramatically in T8 group compared with F8 group.In contrast,the expression of cystatin C mRNA fell to normal levels.Conclusion Cathepsin S,K and cystatin C play an important role in the left ventricular remodeling with pressure overloaded myocardial hypertrophy.
