摘 要:为了摸清重庆地区的菊花品种的遗传关系,以便分类鉴定与生产运用,用ISSR-PCR技术对25个重庆地区主栽菊花品种的遗传多样性和亲缘关系进行了分析。从100条ISSR引物中筛选出10条引物分别对供试材料基因组DNA进行扩增,共扩增出242条带,其中多态性条带241条,多态性比率高达99.6%,表明供试材料在DNA水平上存在广泛差异。用NTSYS软件统计分析出供试品种的DICE相似系数界于0.331-0.718之间,应用根据UPGMA得出聚类结果,将25个品种分成4个类群,聚类结果与传统的形态学分类有些不同,可以看出ISSR标记技术能较准确地揭示出菊花品种的遗传多样性,它与形态学分类方法结合对菊花品种的分类研究会有很大推动作用。ISSR-PCR was used to detect the genetic diversity and relationship of 25 chrysanthemum cultivars.Fingerprinting amplified by 10 ISSR primers revealed a total number of 242 unambiguous bands,of which 241 ones were polymorphic and the polymorphism frequency was 99.6%.There was extensive difference between these varieties at the level of DNA.The similarity coefficient of cultivars was between 0.331~0.718.These 25 chrysanthemum cultivars were divided into four groups by UPGMA(unweighted pair group method with arithmetic average) based on Jaccard coefficient.Results indicated that it is practical to categorize chrysanthemum varieties with ISSRs.The result was different from the traditional morphologic classification methods.So we can reveal the genetic diversity of chrysanthemum cultivars well and truly by ISSR molecular marker,it can push classication research of chrysanthemum forward greatly if we combine morphologic classification methods with molecular marker techniques.
