促生菌对大蒜根尖细胞有丝分裂的影响

摘  要:用不同促生菌(U4,CC9)、促生菌加不同浓度NaCl溶液处理大蒜根尖,并在处理的当天和第2、4、6、8和10天通过常规染色压片技术对大蒜根尖细胞有丝分裂进行观察。结果显示:经过促生菌处理的大蒜根尖细胞有丝分裂指数随时间的延长呈现出明显的升高趋势;用NaCl溶液处理的大蒜根尖细胞有丝分裂指数则有明显的下降趋势,并且诱发产生了染色体桥、染色体粘连、核出芽、不均等分裂、微核等畸变现象;用NaCl和促生菌共同处理大蒜根尖细胞时,随时间的延长有丝分裂指数缓慢升高。提示:促生菌在盐渍化土壤中仍对大蒜根尖细胞有丝分裂具有一定的促进作用,而且时间越长促进效果越明显。The root tip of garlic was treated with different growth promoting bacteria(U4,CC9) and growth-promoting bacteria in different concentrations of NaCl solution,and the mitosis of root tip cells of garlic was observed by conventional dyeing squash technique at the day of treatment and 2,4,6,8 and 10 days post treatment.The results showed that the mitotic index of root tip cells of garlic treated by growth promoting bacteria increased significantly with the passage of time,while the mitotic index treated by NaCl solution decreased significantly,and the distortion phenomenon of chromosome bridge,chromosome sticking,nuclear budding,asymmetric division and micronucleus are induced;the mitotic index increased slowly with the passage of time when the root tip cells of garlic in the co-treatment of NaCl and growth promoting bacteria.The results suggested that the growth promoting bacteria in saline soil still had a promoting effect on the mitotic of root tip cells of garlic,and the longer time was,the more obvious the promoting effect was.
