摘  要:以菹苹的茎为外植体材料,探讨了不同采摘季节及灭菌方式对外植体分化的影响,不同培养基类型对增殖培养的影响,不同激素组合对增殖培养及生根培养的影响。结果表明:菹草采摘的最佳季节为每年3月底至4月初,此时菹草的发芽率高且长出的腋芽强壮,褐化率低;外植体最佳的灭菌方式为3%H2O2(30min)+无菌水冲洗3次+5%乙醇+1.0%oNaClO(15~20Min);液体培养基比固体培养基更有利于菹草的增殖培养,最佳组合为:MS+蔗糖3%+6-BA1.0mg/L+GA3 1.0mg/L;生根培养的最适培养基为:母液I25%+IBA2.0mg/L+蔗糖2%。It is very improtant to restoration and phytoremediation of aquatic plants that tissue culture of Potamogeton Crispus L. was established. The experimental results showed that the optimal way to sterilization fo tissue culture of Potamogeton Crispus L. was 3%H2O2 (30 min)with sterile water washing 3 times, 5% ethanol and 1.0% NaClO(15~ 20 min), and the optimal medium was mother liquor I 25%, IBA 2.0 mg/L and Sucrose 2%. The experiments also showed the results that liquid medium was advantageous to proliferation culture, and the growth speed of adventitious bud of Potamogeton Crispus L was faster than that of solid medium.
