冰片对三氯化铁诱导的大鼠动脉血栓形成的抑制作用及机制

摘　　要：目的:探讨冰片的抗血栓作用。方法:用血栓形成实验、血小板聚集实验及血小板内钙离子浓度的测定,考察冰片对血栓形成、血小板5-羟色胺(5-HT)含量、血小板聚集率及血小板内游离钙离子浓度[Ca2+]i的影响。结果:ig冰片粟米油溶液140 mg.kg-1,7次能抑制三氯化铁诱导的大鼠动脉血栓形成(P<0.05),升高血小板内5-HT含量(P<0.05),抑制5-HT诱导的血小板聚集作用(P<0.05);在有无细胞外钙存在时,冰片血清均能够明显抑制5-HT诱导的血小板胞内[Ca2+]i升高。结论:冰片具有抗血栓作用,其机制可能与抑制血小板5HT释放和血小板聚集,抑制血小板胞浆[Ca2+]i升高有关。Objective：To study on the antithrombotic effect of bornel.Method：Thrombosis test,platelets aggregation,and determination platelet [Ca^2 ＋ ]i were carried out to investigate the effects of borneol on the following indexes：weight of thrombus,the concentration of serotonin in platelets,platelet aggregation rate,and intracellular free calcium concentration in platelets.Result：Ig 140 mg·kg^-1 borneol daily for 7 days inhibited arterial thrombosis induced by FeCI3,increased the concentration of serotonin in platelets,and inhibited the platelet aggregation induced by 5-HT（P〈0.05）;with or without the presence of extracellular Ca^2 ＋,bornel inhibited the increased [Ca^2 ＋ ]i induced by 5-HT.Conclusion：Borneol has anti-thrombosis,and its mechanism may be related to inhibitory effects for the release of platelet 5-HT,for platelet aggregation and for the increase of platelet [Ca^2 ＋ ]i.
