尾加压素Ⅱ与妊娠期高血压疾病发病关系的研究

摘  要:目的探讨尾加压素Ⅱ(UrotensinⅡ,UⅡ)在妊娠期高血压疾病(pregnancy-induced hyertension,PIH)发病中的作用。方法 (1)测定40例PIH患者和16例正常妊娠妇女血清中UⅡ及血钙水平,分析二者之间的相关性。(2)分别利用血清及10^(-7)mol/L的人重组UⅡ(rUⅡ)对离体培养的人脐静脉内皮细胞(HUVEC)作用,同时将硫氮唑酮加入10^(-7)mol/L rUⅡ对离体培养的HUVEC作用。(3)流式细胞仪分析PIH患者血清和rUⅡ在不同作用时间内皮细胞的凋亡率。结果 (1)PIH组孕妇血清UⅡ7.85±0.74pg/mL,血钙1.85±0.35 mmol/L,正常妊娠组血清UⅡ5.21±0.96 pg/mL,血钙2.26±0.18 mmol/L,两组差异均有显著意义(t=2.32,P<0.05;t=2.84,P<0.01),且PIH各组血清UⅡ及血钙水平均呈显著负相关关系(P均<0.05)。(2)正常孕妇血清对培养的HUVEC细胞凋亡率低,PIH患者血清及rUⅡ对培养的HUVEC有同样的促细胞凋亡作用,表现为凋亡率明显升高,两者呈时间依赖性。(3)硫氮唑酮可抑制UⅡ诱导的细胞凋亡。结论 (1)UⅡ可诱导HUVEC的凋亡作用,且这种作用与细胞内Ca^(2+)升高有关。(2)UⅡ可能参与了PIH的发病过程。Objective: To investigate the role of Urotensin Ⅱ ( U Ⅱ ) in the pathogenesis of pregnancy induced hypertension (PIH) by damaging endothelial cells. Methods: 40 patients with PIH and 16 healthy pregnant women were studied. The concentration of U Ⅱ and calcium in maternal plasma were measured, Correlation analysis of them was maded. Endothelial cells were incubated in the presence of rU Ⅱ ( 10^-7mol/L) and sera from PIH women and normal Pregnan. In addition, diltiazem of 400 ng/ml was added into the rU Ⅱ with 10^-7mol/L group, the apoptotic rate was counted in different time periods ( 12 h, 24 h, 48 h, and 72 h) by flow eytometry. Results: Maternal plasma U 11 and calcium level in PIH were 7. 85±0. 74pg/ml and 1.85 ± 0. 35 mmol/L, higher than that in normal pregnancy (thelatter were 5.21 ±0.96 pg/ml and 2. 26±0.18 mmol/L, (t =2.32, P 〈0.05; t = 2. 84, P 〈 0. 01 ). There were negative correlation in PIH. Both sera of PIH women and rU Ⅱ induced the typical apoptosis of HUVEC in a time - dependent manner. Dihiazem might significantly suppress the U Ⅱ - induced apoptosis. Conclusion : U Ⅱ could induce the apoptosis of HUVEC, which may be associated with the increase of [ Ca^2 + ] i. U Ⅱ may be involved in the pathogenesis of PIH.
