摘 要:目的探讨烧伤血清中是否存在内毒素致敏物质及其在内毒素诱导肝细胞损伤中的作用。方法以Wistar大鼠为对象,采用二步灌注法分离肝细胞及枯否细胞,分别与内毒素和烧伤血清培养后,观察培养上清中乳酸脱氨酸(LDH)及肿瘤坏死因子(TNF)的变化。结果烧伤血清可显著提高枯否细胞对内毒素的敏感性,诱导枯否细胞释放LDH及TNF明显高于对照组,同时可显著增加枯否细胞与肝细胞联合培养时LDH水平。结论在烧伤血清中存在内毒素数敏物质,可显著提高机体对内毒素的敏感性,在内毒素浓度较低时,仍可诱导枯否细胞释放TNF,从而损伤肝细胞,其中内毒素结合蛋白sCD14可能起重要作用。Objective To study whether illers exists substance increasing tile sensitivityto endoloxin in burn rat serum and its role ill the endotoxill-induced liver injury. MethodsKupffer cells and hepatocytes were isolated from Wistar rats by a two-step method, andtumor necrosis factor (TNF) and lactate dehydrogenatase (LDH) were detected in the supernatant of Kupffer cells cocultured with hepatocytes. Results The burned rat serum couldsignificantly increase the sensitivity of Kupffer cells to elldotoxin, and increase the concentration of endotoxin in burn serum group, inducing Kupffer cells to release LDH. And the level of TNF was higher than that of the control group. And the burn serum could enhance LDHlevel after hepatocyte injury induced by Kupffer cells. Such phenomenon was not found in thenormal serum group. Conclusion it suggests that there exists substance increasing thesensitivity to endotoxin ill the burned rat serum, which can significantly increase thesensitivity of Kupffer cells to elldotoxin and induce hepatocyte injury by TNF and othermediators released from KuDffer cells. evefl in very low endotoxin concentration.
