摘  要:运用逐因子实验确定生物酶洗毛的最优菌种方式为枯草芽孢杆菌+解脂假丝酵母,其接种比例以1∶4(枯草芽孢杆菌∶解脂假丝酵母)为佳,最适培养条件:温度40℃,pH值7.0,酶用量6%(owf),浴比1∶35,时间16 h。在此基础上,通过Plackett-Burman设计在影响酶法洗毛的5个因素中,有效筛选出主效因素:浴比、温度和时间。在用最陡爬坡实验逼近以上3个因子的最大响应区域后,采用响应面分析法(RSM)对以上3个显著因子的最佳水平范围进行研究,通过对二次多项回归方程求解得知,酶法洗毛最适条件为:浴比1∶33.28、温度40.44℃、时间18.11 h。经洗毛实验验证,洗净毛的含脂率为0.75%,相对于优化前的工艺,纤维含脂率降低了0.31%,证明RSM法优化生物酶洗毛工艺是可行的。Through the single factor experiment,the optimum enzymatic wool scouring conditions were as follows: bacteria selection: Bacillus subtilis + Candida lipolytica,its proportion 1 ∶ 4(Bacillus subtilis∶ Candida lipolytica),temperatur 40 ℃,pH value 7.0,amount of enzyme 6%,liquor ratio 1∶ 35,and reaction time 16 h.On this basis,Plackett-Burman design was undertaken to evaluate the effects of the five factors.By regression analysis,liquor ratio,temperature and time were found to be important for enzymatic wool scouring.In the second phase of the optimization process by steepest ascent,the response surface methodology(RSM) was used to optimize the above critical factors,and to find out the optimal concentration levels and the relationships between these factors.By solving the quadratic regression model equation using appropriate statistic methods,the optimal parameter of the variables were determined as: bath ratio 1 ∶ 33.28,temperature 40.44 ℃,and time 18.11 h.In the optimum condition,the resin content was 0.75%,which has reduced 0.31% compared to that of non-optimized condition.The experiment data under various conditions have validated the theoretical values.
