阿藿烯对血栓形成的影响及其机制

摘  要:目的探讨阿藿烯对血栓形成的影响及其机制。方法将SD大鼠随机分为正常对照组、阳性对照组、阿藿烯低、中、高剂量共5组,灌胃7d后采用动-静脉旁路血栓形成法制作大鼠血栓模型,测量血栓湿重,检测血清P-选择素(Ps)和血管细胞黏附分子-1(VCAM-1)水平;并以花生四烯酸(AA)作诱导剂,观察阿藿烯对兔血小板聚集的作用。结果低、中、高剂量组阿藿烯均可显著抑制血栓形成(P<0.01或P<0.05),显著降低血小板最大聚集率及血清Ps水平(P<0.01);中、高剂量组阿藿烯还可显著减少VCAM-1含量(P<0.01)。结论阿藿烯能够抑制大鼠动-静脉旁路的血栓形成,其机制与减少血小板聚集,抑制VCAM-1和Ps的表达有关。Objective To investigate the effect of ajoene on thrombosis and explore the possible mechanism.Method SD rats were randomly divided into five different groups and administered ajoene in 7d.The effect of ajoene on thrombosis in rats was observed with the thrombogenesis model of artery-vein bypass.The serum level of vascular cell adhesion molecule-1(VCAM-1) and P-selectin(Ps)were measured.To observe the effect of ajoene on platelet aggregation,arachidonic acid was used to induce platelet aggregation and platelet function was evaluated by turbidimetry in rabbit.Results Ajoene significantly inhibited the the thrombosis of artery-vein bypass in rats and reduced rate of platelet maximal aggregation and decreased the P-selectin concentration in serum at 25,50,75mg/kg (P0.05 or P0.01),but VCAM-1 level was only reduced (P0.01) at 50,75mg/kg in rat.Conclusion Ajoene can inhibit the thrombosis in rats.The mechanism may partly be due to decrease of VCAM-1,P-selectin and inhibition of platelet aggregation.
