摘 要:用CTAB法从冬瓜种子发芽幼根提取DNA,经电泳检测其条带清晰明亮,可满足RAPD分析要求。对黑皮冬瓜黑老粗F1及其亲本DNA用140个RAPD引物分别进行RAPD扩增,筛选获得1个在父母本间可产生稳定且为双亲互补型片段的引物SPS-C15。利用该引物对一批商品种进行纯度鉴定,纯度为90.7%,实际田间形态鉴定的纯度为89.8%,RAPD检测结果与田间鉴定结果基本吻合,表明RAPD技术可用于黑老粗F1的种子纯度快速检测。Using CTAB method, template DNAs were extracted from young roots of germinated wax gourd(Benincasa hispida Cogn.)seeds. Electrophoresis revealed that DNA bands extracted were clear and bright, indicating that the DNA quality could suit RAPD analysis. PCR amplification profiles of black rind wax Gourd variety-‘Heilaocu’F1 and its parents were analyzed by using 140 RAPD primers. Only one primer, SPS-C15 was found to generate stable and complementary special DNA fragment. A production seed sample was identified with this primer, showing 90.7 % of seed purity. While the seed purity was 89.8 % in field test. The results of these two methods were consistent, suggesting that RAPD technology could be used for rapid testing of seed purity in wax gourd F1 hybrid.
