摘  要:用DNAStar软件对GenBank中西尼罗热病毒(WNV)的E基因进行序列分析,设计WNV特异性探针并标记生物素,与荧光编码微球偶联后与病毒E基因的PCR产物杂交反应,用液相芯片检测仪(Liquichip 200)检测荧光信号建立WNV快速液相芯片检测方法。结果表明:该法具有较好的特异性,不与其他虫媒病病毒反应;检测灵敏度达到100个拷贝。说明初步建立了检测WNV的液相芯片技术,这为进一步搭建WNV全新快速高通量检测平台奠定基础,也为其他同类病毒的快速高通量检测提供借鉴和经验。The E gene of WNV in the GenEBank was sequenced by using the software DNAStar 5.0. Specific WNV probe labeled with biotin was prepared and coupled with fluorescence-coded microspheres. The probe was used for hybridization reaction to PCR products of WNV, then Liquichip detection technique for detection of WNV was established by using Liquichip 200 to detect fluorescence signal in the reaction system. The results showed that this assay method displayed better specificity to WNV and no respond to other insectborne disease virus when being used for detection. The sensitivity test indicated the detecting limit for the WNV could reach to 100 copy. The Liquichip detection technique to detect WNV was established preliminarily, which provided a foundation for further research on rapidly high throughput detection for WNV and also for exploration to detect other viruses with the technique in poultry industry.
