摘  要:目的确定大黄多糖的最佳提取条件。方法以大黄多糖的含量为指标,以提取温度、提取时间、提取次数、料液比为因素,利用正交试验法进行试验设计,对传统水提-醇沉法提取大黄多糖的工艺进行优化研究。结果大黄粗多糖得率最高的提取条件是:提取时间3h,提取温度95℃,料液比1︰30;大黄粗多糖纯度最高的条件是:提取时间1h,提取温度95℃,料液比1︰10;乙醇沉淀大黄多糖的最佳浓度是80%。结论本方法实验结果可作为提取大黄多糖的工艺制定的依据。Objective To determine the optimum extracting conditions of rhubarb polysaceharide. Methods By orthogonal test, take the content of rhubarb polysaccharide as standards, extraction temperature, extraction time, extraction times and ratio of material to liquid as factors, to optimize and study the process of ex- tracting rhubarb polysaceharide through the traditional water solution and alcohol sedimentation. Results The optimum extraction condition of rhubarb thick polysaeeharide has been established as follows : extraction time was 3.0 hours, extraction temperature was 95 ℃, and ratio of material to liquid 1 : 30. The optimum purity condition of rhubarb thick polysaceharide had been established as follows : extraction time was t. 0 h, extraction tempera- ture was 95 ℃, and ratio of material to liquid 1 : 10. The best density of ethanol depositing rhubarb polysaecha- ride was 80%. Conclusion The experimental result of this method can be the basis of extracting rhubarb poly- saeeharide process.
