芦荟生肌膏精提液对内毒素刺激的成纤维细胞增殖的影响

摘　　要：目的:观察芦荟生肌膏精提液对内毒素刺激的成纤维细胞增殖的影响。方法:在不同浓度的芦荟生肌膏精提液的作用下,通过MTT法检测芦荟生肌膏精提液对内毒素(LPS)刺激的成纤维细胞增殖的影响,并在光镜下观察各组细胞的形态学改变。结果:与对照组比,LPS刺激组形态学观察有明显改变,LPS显著抑制成纤维细胞的增殖(P<0.001);与LPS刺激组相比,加入不同浓度中药精制提取液后,可以不同程度地减轻LPS的抑制作用,成纤维细胞的增殖恢复,各组形态学观察也有相应程度的改变,其中以大剂量组最为明显(P<0.01)。结论:内毒素(LPS)可以显著抑制体外成纤维细胞的增殖,镜下表现为细胞稀疏,生长减慢,细胞形态拉长。芦荟生肌膏精提液对LPS刺激的成纤维细胞增殖有保护作用,可以拮抗LPS的抑制作用,恢复成纤维细胞的增殖,提示芦荟生肌膏可以通过促进成纤维细胞的增殖而促进直肠创面组织的愈合。Objective To investigate the influence of Luhui Shengji（芦荟生肌） ointment on the proliferation of fibroblasts stimulated by lipopolysaccharide. Methods After using different concentration of the extract of LuHui Shengji ointment, the proliferation of fibroblasts stimulated by lipopolysaccaride was measured by MTT analysis. The morphologic changes of fibroblasts of each group were examined under microscopy respectively. Results Compared with control group there was significant difference in the morphology of fibroblasts in groups stimulated by lipopolysaccaride. Moreover, LPS significantly inhibited regeneration of fibroblasts （P 0.001）. Compared with the group stimulated by lipopolysaccaride after using different concentrations of the extract of LuHui Shengji ointment , the proliferation and the morphology of fibroblasts of each group was enhanced coinstantaneous. Further, that of the large dose group was most presentive （P 0.01）. Conclusion （1）LPS endotoxin has the effect of inhibition on proliferation of fibroblasts. （2） LuHui Shengji ointment can promote the proliferation of fibroblasts by inhibiting the negative effect of LPS on the proliferation of fibroblasts, hence LuHui Shengji ointment can promote anorectal wound healing
