健康汉族人外周血CD4^+CD8^+双阳性和CD4^-CD8^-双阴性T淋巴细胞的分布

摘　　要：目的:观察健康汉族人外周血中CD4+CD8+双阳性(double positive,DP)T细胞和CD4-CD8-双阴性(double negative,DN)T细胞的分布频率。方法:利用三色流式细胞术分析36例健康成人(<65岁)、34例健康老年人(≥65岁)和22例免疫抑制者外周血DP T细胞和DN T细胞的分布。结果:老年组外周血DP T细胞比例1.93%±1.52%显著高于免疫抑制组1.23%±0.81%(P=0.05);但与成人组1.54%±1.24%相比,老年组外周血DP T细胞比例虽然有增高的趋势但未见显著差异。老年组、成人组和免疫抑制组外周血DP T细胞绝对计数分别为(0.028±0.022)×109细胞/L、(0.024±0.017)×109细胞/L和(0.021±0.019)×109细胞/L,三组间差异无显著性。成人组外周血DN T细胞比例为8.81%±5.56%,明显高于老年组的5.52%±3.49%和免疫抑制组的3.72%±1.21%(P值分别为0.012和0.000);老年组DNT细胞比例也高于免疫抑制组(P=0.025)。老年组、成人组和免疫抑制组外周血DN T细胞绝对计数分别为(0.083±0.069)×109细胞/L、(0.150±0.111)×109细胞/L和(0.057±0.027)×109细胞/L,成人组外周血DN T细胞的绝对数明显高于老年组(P=0.011)及免疫抑制组(P=0.000),而免疫抑制组与老年人比较则未见显著差异(P=0.155)。成人组外周血更常出现CD3分子表达增高(CD3high)的T细胞。与CD3分子表达正常(CD3norm)的T细胞相比,CD3highT细胞群体中包含大量DN T细胞。结论:年龄≥65岁的健康汉族老年人外周血DP T细胞未见明显增多,但外周血DN T细胞随着年龄增高而明显减低。Objective：To analyze the distributional frequency of peripheral CD4＋CD8＋ double positive（DP） and CD4-CD8-double negative（DN） T cells in healthy Han people.Methods：The distribution and absolute count of peripheral DP T and DN T cells in 36 healthy adults aged less than 65 years（adult group）,34 healthy elderly aged 65 years or more（elderly group）,and 22 immunocompromized patients（immunosuppressed group） were determined with tri-color flow cytometry and Flow Count beads.Results：The percentages of circulating DP T cells in elderly groupa,dult group,and immunosuppressed group were 1.93%±1.52%,1.54%±1.24%,and 1.23%±0.81%,respectively.A marginal significant differ-ence could be observed on the frequency of peripheral DP T cells between elderly group and immunosuppressed group（P=0.05）,there was only a tendency to accumulate in peripheral blood for DP T cells in elderly but no significant difference was observed between elderly and adult group.What’s more,there was no any significant difference on the absolute count of DP T cells among the three groups with absolute counts of（0.028±0.022）×109 cells/L（elderly）,（0.024±0.017）×109 cells/L（adult）,and（0.021±0.019）×109 cells/L（immunosuppressed）,respectively.The frequency of DN T cells in elderly（5.52%±3.49%） and immunosuppressed group（3.72%±1.21%） decreased significantly when compared with adult group（8.81%±5.56%,P values were 0.012 and 0.000,respectively）.There was also a significant difference between elderly group and immunosuppressed group（P=0.025）.The absolute counts of peripheral DN T cells in elderly,adult and immunosupressed group were（0.083±0.069）×109 cells/L、（0.150±0.111）×109 cells/L and（0.057±0.027）×109 cells/L,respectively,with sig-nificant differences observed between elderly group and adult group（P=0.011）,between immunosup-pressed group and adult group（P=0.000）,respectively.Compared with CD3＋ cells with normal CD3molecule density（CD3norm）,a smal
