摘  要:目的:研究卵泡抑素在爪蟾心形成过程中的作用。方法:切下发育第14期胚胎形成心的组织,并分别加入卵泡抑素、骨形成蛋白和活化素进行培养。4h时后,用MEMPHA固定并进行胚胎整体原位杂交。利用Morpholio Oligo技术减弱了卵泡抑素的表达,检测了在缺乏卵泡抑素条件下心形成的变化。结果:早期心表达卵泡抑素;增加或降低卵泡抑素均抑制Nkx2.5的表达。结论:卵泡抑素在心的形成过程中发挥十分重要的作用,缺乏或过量卵泡抑素均影响心的形成。Objective: To investigate the role of follistatin in the heart formation in xenopus laevis. Methods: Explants from embryos of xenopus laevis at stage 14 were isolated for culture with follistatin, BMP and activin. Four hours later, RNA tran- scripts were examined by whole mount in situ hybridization. For morpholino experiments, embryos were injected at one-ceil stage with 5 or 10 ng of foltistatin-MO or control MO solution. At stage 20, RNA transcripts were examined by whole mount in situ hybridization. Results: Follistatin was expressed in the early heart as well as in the neural tissues; Either overexpression of follistatin or deficient follistatin inhibited Nkx 2.5 expression in the heart explants. Conclusion: Follistatin is a key player in heart development and plays an essential role in regulating heart formation in xenopus laveis.
