摘 要:目的通过观察运动配合左归丸对去卵巢大鼠骨质疏松的偶联信号因子骨髓基质细胞IL-1,成骨细胞IL-6、COX-2的影响,探讨该方法治疗骨质疏松症的作用机理。方法随机分为正常对照组、假手术组、模型组、己烯雌酚组、运动组、左归丸组、运动配合服用左归丸组。采用免疫组化的方法观察胫骨中白细胞介素1(IL-1β)、白细胞介素6(IL-6)和环加氧酶2(COX-2)3种细胞因子的蛋白表达,并运用ELISA技术测定血清中雌二醇(E2)、降钙素(CT)、骨钙素(BGP)、甲状旁腺激素(PTH)含量。结果大鼠造模后,胫骨IL-1、IL-6、COX-2蛋白表达显著增高,血清中CT、E2显著性降低,BGP、PTH显著性升高。运动配合服用左归丸可以对升高的IL-1、IL-6、COX-2产生明显的抑制作用,且运动配合服用左归丸组与运动组相比IL-1β、IL-6有显著性差异,与左归丸组相比IL-1β、COX-2有显著性差异。与模型组相比,运动配合左归丸组CT、E2升高,BGP、PTH均降低,且运动配合左归丸组血清E2较左归丸组及运动组有显著性升高。结论运动配合左归丸可使去卵巢大鼠体内雌激素分泌增加,进而抑制骨质疏松偶联信号因子IL-1β、IL-6、COX-2分泌,间接地抑制骨吸收,调节骨吸收与骨形成的动态平衡,达到治疗绝经后骨质疏松症的目的。Objective To study its mechanism on the treatment of Osteoporosis based on the effect of movement combined Zuogui Pills on the osteoporotic couple signal transmission factors of ovaries extirpation rats, marrow stroma cell (MSC) IL-1, osteoblast IL-6 and COX-2. Methods Bilateral ovaries extirpation osteoporotie rats were divided into 7 groups, negative control group, sham operation group, model group, Diethylstilbestrol group, movement group, Zuogui Pills group and movement combined Zuogui Pills group. The protein expressions of three cytokines, interleukins (IL-1β, IL-6) and cycloxygenase cyclo-oxygenase (COX-2) of the tibia had been observed by histogenic immunohistochemistry, and the contents of estradiol ( E2 ) , Calcitonin (CT) , bone gla protein (BGP) and parathyroid hormone ( PTH ) of the blood serum had been assessed by ELISA. Results The protein expressions of IL-1, IL-6 and COX-2 of the tibia of the ovaries extirpation rats increased obviously, but the contents of serum CT and E2 decreased significantly, while the contents of serum BGP and PTH increased obviously. The therapy of movement combined Zuogui Pills had a significant inhibition on the increased protein expressions of IL-1 , IL-6 and COX-2. Compared with those of pure movement group, protein expressions of IL-1β and IL-6 of the combination treatment group had significant differences. Compared with those of pure Zuogui Pills group, protein expressions of IL-1β and COX-2 of the combination group had significant differences. The contents of CT and E2increased while BGP and PTH decreased in the combination treatment group. The group comparison showed that there were not any differences among the pure Zuogui Pills group, pure movement group and the combination group except that the serum E~ in the combination group was higher than those of the other 2 groups. Conclusion The combination of movement and Zuogui Pills could promote the estrogen secretion of ovaries extirpation rats, then inhibit the secretion of osteoporo
