摘  要:Mitf(Microphthalmia-associtated transcription factor)是小眼畸形相关转录因子,在黑色素细胞的发育、分化和功能调节上起着重要作用,同时还参与了其它3种细胞包括肥大细胞、破骨细胞和眼色素上皮细胞的发育和分化。针对猪的Mitf基因,从GenBank核酸数据库和proenseml网站下载4条猪的含有Mitf基因片段的基因组序列与牛、人、鼠3种物种的Mitf基因组序列进行比较基因组分析,结果发现,下载的猪Mitf基因组序列或没有排序,或存在一定错误排列。通过生物信息学和比较基因组分析,将猪Mitf基因的基因组序列进行重新组装,得到由6条有序基因组片段组成的较为完整的Mitf基因组序列,并根据人类Mitf基因的转录本信息推测出5种不同猪Mitf基因的转录本。进化分析表明,在人类、小鼠、猪、牛、马和狗中,猪和牛Mitf基因的进化关系最近,提示做猪基因的生物信息学分析可借鉴牛的基因分析结果。Mitf( Microphthalmia-assoeitated transcription factor)plays an important role in development,differetiation and functional regulation of melanocytes and three other cells including mast cells,osteoclasts.and retina pigment epithelial cells. In this paper, 4 genomic sequences containing the fragments of pig Mitf gene, downloaded from GenBank nucleoide database or proensembl website, were compared with cattle,human and mouse Mitf genomic sequences that were downloaded from UCSC genome browser. The aligment results showed that these 4 porcine genomic sequences were unordered or misarranged. Therefore,based on the results of comparative genonic analysis, porcine Miff genomic sequences were rearranged into 6 ordered fragments in silico. 5 different pig Miff transcripts were predicted compared with human Mitf gene transcripts. Phylogenetic analysis suggested that the evolutionary relationship of Miff gene between pig and cattle were closer than the one between pig and human, mouse, horse or dog. It inferred that the results of bioinformatic analysis for cattle genes could be good referrence for pig genes.
