摘 要:采用放射配基结合法,利用GraphPad PRISM5.01软件计算出受体及其亚型的密度,从而比较α1肾上腺素能受体(α1-AR)亚型在大鼠输尿管各段中的分布情况。α1-AR在大鼠输尿管各段的结合位点是非均匀分布的,下段输尿管的Bmax为(14911±1253)pmol/g,明显高于上段(9077±211)pmol/g及中段(9164±158)pmol/g,上段与中段的分布差别不显著。粗制膜标本经过CEC预温育后,α1-AR与[125I]-HEAT的最大结合容量Bmax比对照组下降约64%。在有α1-AR亚型高特异性拮抗剂(5-MU,Cyclazosin,BMY7378)存在时α1-AR与[125I]-HEAT的最大结合容量Bmax均有不同程度的下降。放射性配基结合试验结果显示,α1-AR在输尿管下段的密度要高于上段及中段,但Kd值及Hill系数并没有区别。通过单点竞争抑制反应表明,大鼠输尿管中存在3种α1-AR亚型,即α1A-AR、α1B-AR和α1D-AR亚型,但是α1D-AR亚型与α1A-AR亚型的分布要明显多于α1B-AR亚型,而α1D-AR亚型分布略高于α1A-AR亚型。The distribution characteristics of α1-adrenoceptors(a1-AR) subtypes in proximal,medial,and distal regions of the rat ureter were investigated.The experiment was studied by using the radioligand binding assay and GraphPad PRISM 5.01 software to analyze the data,calculate the density of α1-adrenoceptors(a1-AR) subtypes,and compare the distribution of subtypes in the different regions of the rat ureter.Binding sites of α1-AR in the different regions of the rat ureter were non-uniform distribution,the Bmax for lower ureteral was (14 911±1 253) pmol/g,significantly higher than the proximal (9 077±211) pmol/g and medial (9 164±158) pmol/g,whereas the distribution difference between the proximal and medial was not significant.Compared with the control group,the maximum binding capacity (Bmax) of α1-AR and [125I]-HEAT decreased about 64% in the crude membrane specimen through the CEC pre-incubation.When the high specificity antagonist of α1-AR subtypes (5-MU,cyclazosin and BMY7378) were present,Bmax of α1-AR and [125I]-HEAT were decreased.Radioactive ligand binding experiments showed that α1-AR density in the distal ureter was more than that of the proximal and medial,but there was no difference between the Kd and Hill coefficients.A single point of competitive inhibition reaction showed that three kinds of α1-AR subtypes existed in the rat ureter,namely,α1A-AR,α1B-AR and α1D-AR subtypes,but α1D-AR subtypes and α1A-AR subtypes were significantly more than α1B-AR subtype,while the distribution of α1D-AR subtype was slightly more than α1A-AR subtype.
