摘  要:以秦皮为主药,瞿麦为辅药,探讨纯中药制剂抗炎作用机制。选用Wistsar种清洁级雄性大白鼠60只,体质量(216±2.8)g,随机分为4组,除空白对照组外,用10%乙酸造成溃疡性结肠炎(Ulcerative colitis,UC)。采用酶联免疫分析法(ELISA)测定了正常、造模及治疗后1、3、7d大白鼠血清中前列腺素E2(Prostandin,PGE2)、白介素-1(interleukin-1,IL-1)和肿瘤坏死因子(tumor necrosis factor,TNF-α)的水平;同时,采用放射免疫分析法检测了UC模型大鼠结肠黏膜组织中环磷酸腺苷(Cyclic adenosine monophosphate,cAMP)含量的变化,探讨抗炎作用机制。结果显示,在大鼠患UC时,血清中PGE2、IL-1和TNF-α明显升高,黏膜组织中cAMP显著降低(与空白对照组比较,P<0.01);该方治疗后,大鼠UC模型血清中PGE2、IL-1和TNF-α的含量显著降低,cAMP含量显著升高(与模型对照组比较,P<0.01)。结果表明,该方剂抑制结肠黏膜组织促炎因子的释放和提高cAMP含量是其抗炎作用机制之一。This artical reports the antinflammation function of Tibet Qinqu medicament(Qinpi as the main therapy and mixed with Qumai to a certain percentage).60 wistsar male healthy big rats(weighted (216±2.8) g each) were divided into 4 group at random: the normal control group(A),the model control group(B),the traditional medicine treatment gruop(C),the positive control group(D).Except for group A,the other three groups were inflammatory induced by 10% acid to ulcerative colities (UC).Using the ELISA measured and analyzd the changes of the contents of interleulin-1(IL-1),tumor necrosis factor-α(TNF-α) and prostaglandin E2 (PGE2) in big rats serum.Blood serum are tested before and after the inflammatory,and also tested at the 1st,3rd and 7th days after the treatment.Meanwhile radioimmuoassay mothod test the changes of content of cyclic adenosine monophoshate(cAMP) in big rates colon tissues to analyze the mechanism of treatment for the coliform bacterial K88 in yeallow or white scour of piglets.The test shows that the content of PGE2,IL-1 and TNF-α in blood serum increases as well as the content of cAMP decreases in colon tissue(compare with group A,P0.01) after UC is induced.With the treatment of Tibet Qinqu medicament,the contents of PGE2,IL-1 and TNF-α in blood serum decreases and the content of cAMP increases in colon tissue(compare with group B,P0.01).The result indicates that the mechanism of anti-inflammation of the Qinqu medicament is related to cell factors.
