摘  要:目的从马齿苋(portulaca oleracea L.)中分离、制备总黄酮,用紫外分光光度法测定其含量。方法用含3%(g/ml)氢氧化钠和60%(ml/ml)乙醇碱性醇液煎煮提取总黄酮,聚酰胺柱树脂柱层析分离纯化。以芦丁为对照品,以亚硝酸钠-硝酸铝-氢氧化钠为显色剂,用分光光度计在510nm可见光处测定黄酮含量。结果芦丁标准品溶液浓度在8~48mg/L范围内与吸光度有良好线性关系,回归方程为Y=0.005X-0.001,r2=0.9998,精密度实验RSD为0.69%,稳定性实验RSD为2.04%,重复性实验RSD为2.1%,平均回收率为98.55%,RSD为0.58%,马齿苋全草中总黄酮的含量为7.67%。结论本法测定马齿苋总黄酮的结果准确,可靠,能用于测定马齿苋全草中总黄酮的含量。Objective To separate and purify total flavonoids from portulaca oleracea L. and determinate its content by UV spectrophotometry. Methods The total flavonoids from portulaca oleracea L. were extracted by 60% ethanol and 3 % sodium hydroxide. Then separated and purified by polyamide resin chromatography. Based on reference substance of rutin, with NaNO2-AI(NO3)3-NaOH as chromogenic agent, the determination was performed by UV spectrophotometry at the wave length of 510 nm. Results The absorbency and concentration of ru- tin have good linear correlation in the range of 8-48 mg/L. The regression equation was Y= 0. 005X-0. 001 (r^2 = 0. 999 8). Accuracy experiment RSD=0. 69% ,stability test RSD=2.04% ,repeatability test RSD=2. 1% ,the average recovery rate was 98.55%,RSD= 0. 58%. The content of total flavonoids in portulaca oleracea L. was 7. 67%. Conclusion The method is simple and fast with good stability,accuracy and repeatability. It can be applied to determinate the content of total flavonoids in portulaca oleracea L.
