摘 要:采用AFLP技术对辣椒细胞质雄性不育系21A及其相应的恢复系湘紫基因组DNA进行多态性比较,筛选了64对AFLP选择性引物PstⅠ-NNN+MseⅠ-NNN组合,有58对引物组合在两系间扩增出5156条带,多态性位点为4个,主要表现为条带数目、条带位置和条带强弱的差异。其中在恢复系材料中仅有3个多态性片断,找到了辣椒细胞质雄性不育恢复系湘紫基因组DNA特异的AFLP片段ACA/CGC300、AGT/GGT375和AGC/CGC380,并对这些特异片段的来源及其在细胞质雄性不育恢复系中的作用进行了讨论。AFLP technique with 64 pairs of primer combinations Pst Ⅰ-NNN + Mse Ⅰ-NNN were employed to find the polymorphisms between the genomic DNAs of cytoplasm male sterile line 21A and its restoring line Xiangzi in hot pepper.It was shown that 5156 bands were amplified in two lines with 58 pairs of primer combinations.There were 4 polymorphic loci in two lines,and their differences appeared in the number,location and depth of bands.Only 3 polymorphic fragments were found in cytoplasm male sterile restoring line.These specific AFLP fragments of the genomic DNAs of the restoring line were ACA/CGC300,AGT/GGT375 and AGC/CGC380.And the source of these fragments and their effect on the cytoplasmic male sterile restoring line were also discussed.
