摘  要:目的探讨N,N-二甲基甲酰胺(N,N-di methylformamide,DMF)对ICR小鼠神经行为功能的影响。方法 80只ICR小鼠(雌雄各半)随机分为4组,分别以1.26g/kg、0.63g/kg、0.32g/kg及生理盐水每日经口灌胃。染毒前、染毒50d和90d分别进行morris水迷宫、避暗箱、穿梭箱和自主活动箱等4项神经行为功能试验。结果 morris水迷宫和避暗箱试验结果在染毒前、后的各剂量组间均无统计学差异。穿梭箱试验:染毒90d,雌性小鼠高剂量组较对照组电击时间延长,主动逃避时间缩短,存在剂量-效应关系。自主活动箱试验:染毒50d,雄性小鼠高剂量组较对照组自主活动次数增加,存在正性剂量-效应关系;染毒90d后,雄性小鼠高、中、低剂量组的自主活动次数均较对照组减少,存在负性剂量-效应关系。结论 DMF亚慢性染毒对ICR小鼠空间学习记忆能力及短期记忆过程没有影响;对雌性小鼠的条件刺激反应及相关记忆能力有抑制作用,对雄性小鼠中枢神经系统兴奋性的影响表现为先兴奋,后抑制的特点;DMF对ICR小鼠部分神经行为功能的影响存在性别差异和剂量-效应关系。Objective To explore the effect on neurobehavior function when subchronic exposure to N,N-dimethylformamide(DMF)in ICR mice.Methods 80 ICR mice(40 males and 40 females)were divided into 4 groups randomly and treated with DMF through intragastric administration at 1.26,0.63,0.32 g/kg/d and normal saline respectively.Before and after 50,90 days of treatment,the mice's behaviors were tested by Morris water maze test,avoiding dark box test,shuttle-box test,and autonomic activity test.Results There was no statistical difference in test results among the 4 groups either before treatment or after treated for 50 or 90 days in Morris water maze test and avoiding dark box test.After treated for 90 days,the electric shock phase prolonged and the active escape phase shortened in highest-dose group when contrasted with the control group and there was a dose-response relationship between dose and result in female mice in shuttle-box test.In autonomic activity box test,the number of autonomic activities increased after treated for 50 days and decreased after treated for 90 days in highest-dose group when contrasted with the control group in male mice.And there was also a dose-response relationship between dose and the result.Conclusions Subchronic DMF exposure may have no effect on learning and memory function of space and impermanent memory in ICR mice model.But it can inhibit the conditioned stimulus reaction and related memory of female mice and affect the excitement status of central nervous system,which is characterized as being excited first and then depressed in male mice.
