摘  要:目的体外观察异基因脂肪干细胞(adipose-derived stem cells,ADSCs)对活动期系统性红斑狼疮(systemic lupus erythematosus,SLE)患者Th17细胞亚群的影响。方法原代培养人脂肪干细胞,流式细胞仪检测表面标记,进行成脂、成骨诱导。密度梯度离心法分离10例活动期SLE患者外周血的单个核细胞(peripheralblood mononuclear cell,PBMC),分组与第3代或第8代ADSCs共培养,48 h后流式细胞仪检测各组Th17淋巴细胞比例,ELISA检测培养上清中IL-17的含量。结果体外成功培养、鉴定脂肪干细胞。与对照组相比,两个与第3代(P3)ADSCs共培养组Th17比例及IL-17浓度下降,而与第8代ADSCs共培养组却出现相反结果;直接共培养组比Transwell隔离共培养组Th17比例及IL-17浓度低(P<0.05)。结论低代数ADSCs可下调活动期SLE患者外周血Th17细胞比例,抑制其分泌IL-17,而高代数的ADSCs具相反作用。ADSCs对Th17的抑制作用与细胞间接触抑制有关。Objective To investigated the effects of allogeneic adipose-derived stem cells(ADSCs) on Th17 lymphocytes from active SLE patients in vitro.Methods ADSCs obtained from adult subcutaneous fat were cultured in vitro,verified with flow cytometry(FCM),and induced into two lineages.Peripheral blood mononuclear cells(PBMCs) were isolated from 10 cases of active SLE patients by density gradient centrifugation.PBMCs from each patient were divided into 3 coculture groups(with ADSCs of passage 3 or 8) and one control group.Th17 ratio and concentration of IL-17 were measured by FCM and ELISA,respectively,at 48 h.Results ADSCs from primary culture presented fibroblast-like morphology under optical microscope.Cell surface phenotype was consistent with reports.Adipogenic and osteogenic differentiations were successfully induced in vitro.There significantly decrease and increase of Th17 ratio and IL-17 in P3-coculture groups P8-coculture group(P〈0.05),respectively,compared with in control group.In P3-coculture groups,Th17 ratio and IL-17 in P3-coculture group were significantly lower than those in P3-Transwell coculture group(P〈0.05).Conclusion P3 ADSCs can down regulate the amount and capacity of Th17 in SLE patients,while P8 ADSCs per contra.The immunosuppression is partly conducted by cell-cell contact.
