珠美海棠叶片总RNA提取方法的比较

摘　　要：珠美海棠(Malus zumi)为多年生木本植物,组织中含有大量的多酚、多糖和蛋白质等物质,给总RNA的提取带来困难。本试验使用RNeasy Plant Mini Kit试剂盒、Trizol法、SDS法和改良CTAB法提取珠美海棠叶片总RNA,并用凝胶电泳和RT-PCR技术比较了4种不同方法提取的总RNA样品的质量,认为提取珠美海棠叶片总RNA的最佳方法为改良CTAB法。Malus zumi is a woody perennial plant,and the phenolic compounds,polysaccharides,proteins are the main interference factors for the isolation of RNA from M.zumi tissues.Using RNeasy Plant Mini Kit,and the methods of Trizol,SDS and improved CTAB,the total RNA was isolated from the leaves of M.zumi.Tested by agarose gel electrophoresis and RT-PCR test,the results show that the improved CTAB method could isolate high quality total RNA.So the improved CTAB method is the optimum method for isolating total RNA from M.zumi leaves.
