摘 要:目的应用琼脂糖稀释法测定奈瑟淋球菌(NG)药物敏感性,并建立体外NG头孢曲松耐药株。方法配置含有不同浓度的四环素、环丙沙星、壮观霉素、头孢曲松培养基并接种一定量的NG菌悬液,培养24h后观察药敏试验结果。采用头孢曲松次抑菌浓度对NG敏感株体外反复诱导。结果 NG诱导前四环素、环丙沙星、壮观霉素、头孢曲松最小抑菌浓度(MIC)值分别为16μg/ml、16μg/ml、16μg/ml、0.016μg/ml。经过连续120次诱导传代,NG对四环素、环丙沙星、壮观霉素、头孢曲松MIC值分别为32μg/ml、16μg/ml、16μg/ml、1.00μg/ml,耐药子代经过30d的无药传代后,MIC值无明显变化。结论琼脂糖稀释法是测定奈NG药物敏感性的有效、实用的方法 ;通过体外人工诱导可获得稳定性好的NG耐头孢曲松株。Aim To determine the antibiotic susceptibility establish ceftriaxone resistant Neisseria gonorrhoeae (NG) strain and test its snesitivity to aitibiotics by agarose dilution method. Methods Agarose disks were inoculated with NG, and tetracycline,ciprofloxacin,spectinomycin and ceftriaxone were added to the medium in various concentrations. Results The minimum inhibitory concentrations(MICs)of tetracycline,ciprofloxacin,spectinomycin and ceftriaxone before inducement were 16μg/ml,16μg/ml,16μg/ml and 0.016μ?g/ml,respectively. The clinical strain of NG was induced successfully with ceftriaxone after cultured for 120 passages and MICs of tetracycline,ciprofloxacin,spectinomycin and ceftriaxone were increased to 32μg/ml,16μg/ml,16μg/ml and 1.00μg/ml,respectively. The strain was transferred 30 passages on ceftriaxone-free medium and the MICs were the same as those after induced. Conclusion Agarose dilution method is an efficient and practical method for determination of susceptibility of NG to antibiotics and the induction experiment could induce NG acquiring stable resistance to ceftriaxone.
