摘 要:利用梯度筛选法,在Pb2+浓度为1 100 mg/L时从被原铅锌矿区尾矿库污染的土壤中筛选出了一株耐铅细菌。对此株细菌的初步鉴定和培养条件研究显示:该细菌为革兰氏阴性好氧菌,最适摇床速率为150 r/min;对环境温度较敏感,最适培养温度为28~32℃;耐碱性环境,对酸性环境很敏感,最适pH为7.0~7.7。对该株细菌干、湿两种状态下的铅吸附特性分析结果表明:干菌的吸附量始终大于湿菌。干菌和湿菌的吸附量都会随着Pb2+初始浓度的增加而增加;特定初始浓度下吸附量随菌量的增加而减小,干菌大于0.4 g,湿菌大于0.8 g时,吸附量趋于平缓。A lead-tolerant bacteria strain was isolated from the contaminated soil of the tail store of a lead-zinc mine by gradient screening technique when the concentration of Pb^2+ was 1100mg/L.The strain's culture conditions were examined and a preliminary identification was performed.The strain was aerobic,gram-negative,the optimum rotating-bed culture speed of which was 150r/min.The strain was sensitive to ambient temperature with the optimum temperature being 28 ℃-32 ℃;it was alkali tolerant but acid sensitive,with the optimum pH ranging between 7.0-7.7.The lead adsorption characteristics of the strain at dried and wet conditions were analysed.The adsorption capacity at the dried condition was higher than that at the wet condition.The adsorption capacity increased with the increase of Pb2+ initial concentration at both wet and dried conditions.The adsorption capacity decreased with the increase of the amount of bacteria at a certain initial concentration;the adsorption capacity maintained when the amount of dried bacteria was over 0.4 g or the amount of the wet bacteria was over 0.8 g.
